高效液相色谱法同时测定青蒿绿原酸、隐绿原酸、东莨菪内酯含量

摘要:
[目的] 通过高效液相色谱法测定青蒿中绿原酸、隐绿原酸和东莨菪内酯的含量,并对人工规范化种植的青蒿中绿原酸、隐绿原酸和东莨菪内酯进行测定, 为青蒿药材质量评价提供分析方法。[方法] Agilent Zorbax SB-C18反相色谱柱(4.6 mm×250 mm, 5 μm), 乙腈-0.1%甲酸水为流动相, 波长325 nm, 柱温30℃, 流速1.0 mL/min, 进样量10 μL.[结果] 绿原酸、隐绿原酸、东莨菪内酯在范围内呈现良好的线性关系, 方法学验证结果均符合相关要求。[结论] 该方法简便可行, 重复性好, 专属性强, 可用于青蒿药材的质量控制。

Abstract:
[Objective] A sensitive method of the determination of chlorogenic acid, cryptochlorogenic acid and scopoletin content in traditional Chinese medicine Artemisia annua L. was established by high performance liquid chromatography (HPLC). [Methods] The separation was achieved by use of C18 analytical column (4.6 mm×250 mm, 5 μm) at 30℃, while the injected sample volume was set to 10 μL. A constant flow rate of 1.0 mL/min was employed throughout the analysis. A binary gradient elution program of aqueous formic acid (0.1%, v/v) versus acetonitrile was selected for the quantitative analysis of chlorogenic acid, cryptochlorogenic acid and scopoletin. [Results] The HPLC method showed the satisfactory linearity, and the results of the method validation meeted the relevant requirements. The method had been successfully applied to the determination of chlorogenic acid, cryptochlorogenic acid and scopoletin content in Artemisia L. samples. [Conclusion] The established HPLC method could be used to evaluate the quality of Artemisia annua L.
