芪参益气组方对心肌梗死大鼠心肌细胞凋亡和胶原容积的影响

摘要:
[目的] 探讨芪参益气组方对心肌梗死(MI)大鼠心肌细胞凋亡、梗死面积和心肌组织胶原容积分数的影响.方法]成功建立大鼠MI模型后随机分为假手术组(10只),单纯MI组(19只)、MI后芪参益气组方干预治疗组14只).干预4周后测量心肌梗死面积、心肌细胞凋亡指数(MAI)、心肌细胞Bcl-2和Bax蛋白表达、心肌组织胶原容积分数(CVF)等指标.[结果] 与假手术组相比,MI各组Bcl-2蛋白表达水平均明显降低(P<0.05),Bax蛋白表达水平、CVF增高(P<0.01).与单纯MI组相比,芪参益气组方干预组Bcl-2蛋白表达水平升高(P<0.05),梗死面积、MAI、Bax蛋白表达水平、CVF降低(P<0.05).[结论] 芪参益气组方通过抑制凋亡、抗心肌纤维化等作用来减轻心肌梗死后的心室重构,从而保护心肌和改善心功能.

Effects of Qishengyiqi prescription on cardiomyocyte apoptosis and collagen volume fraction in rats with myocardial infarction

Abstract:
[Objective] To study the effects of Qishengyiqi prescription on cardiomyocyte apoptosis, myocardial infarction size and collagen volume fraction in rats with myocardial infarction (MI).[Methods] The rats with myocardial infarction were randomized into 3 groups: the groupA (sham operation with thorax opening without coronary artery deligation, n=10),the groupB (MI only, n=19), the groupC [n=14, treated with Qishenyiqi extractum, 100 mg/(kg·d)×28 after AMI, n=14].After treatment for 4 weeks,the infarct size, myocardial apoptosis index, mean density (MD) of Bcl-2, Bax protein and the collagen volume fraction were monitored.[Results] MD of Bcl-2 protein in the group B and C were more weaker than those of groupA (P<0.05),MD of Bax protein, the collagen volume fraction in the group B and C were stronger than those of group A.MD of Bcl-2 protein in the group C was stronger than those of the group B (P<0.05), the infarct size, MAI, MD of Bax protein and the collagen volume fraction were minor than those of the group B (P<0.05).[Conclusion] Qishenyiqi can relieve the left ventricular remodeling, improve the cardiac function and protect the heart in rats with myocardia infarction by inhibiting myocardium apoptosis and myocardial fibrosis.Key words
