[目的] 探讨黄芪多糖对心肌缺血再灌注损伤的保护作用及其可能的机制。[方法] 48只SD大鼠随机分为假手术组、模型组、黄芪多糖低剂量组(300 mg/kg)和黄芪多糖高剂量组(600 mg/kg)。采用冠状动脉结扎法建立大鼠心肌缺血再灌注损伤模型。观察心肌形态学变化,测定心肌梗死面积,血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)含量,和心肌组织中p65, IκB表达。[结果] 黄芪多糖预处理可改善缺血再灌注损伤引起心肌组织的形态学变化,降低心肌梗死面积,降低p65表达,增加IκB 表达。[结论] 黄芪多糖对缺血心肌再灌注损伤具有一定的保护作用,其机制可能通过下调核因子-κB(NF-κB)通路,降低炎症反应有关。 |

Abstract: |
[Objective] The aim of the present study was to investigate the protective effect of astragalus polysaccharides APS on myocardial ischemia reperfusion injury and the underlying mechanism. [Methods] The 48 SD rats were randomly divided into 4 groups: sham operation group (Sham group), myocardial ischemia-reperfusion (MI/R) group, APS (300 mg/kg) group and APS(600 mg/kg) group. Ischemia was produced by ligating the left anterior descending coronary artery. Myocardial infarct size was determined; myocardial pathological changes were observed, TNF-α, L-6, p65 and IκB protein expression was determined. [Results] Pretreatment of APS before MI/R could attenuate the morphological change of myocardial tissue, decreased the infracted sized, the content of TNF-αand IL-6, p65 protein content, and increased the IκB protein expression. [Conclusion] Astragalus polysaccharides pretreatment could attenuate MI/R injury through inhibition of the NF-κB signaling pathway, which lead to reduced inflammatory reaction. |
