通脉养心丸对异丙肾上腺素所致大鼠心肌肥厚的影响

摘要:
[目的] 观察通脉养心丸对异丙肾上腺素(ISO)所致大鼠心肌肥厚的影响。[方法] 健康雄性Wistar大鼠随机分为6 组:空白对照组,心肌肥厚模型组、通脉养心丸2.0、1.0、0.5 g/kg 组和倍他乐克组。ISO 5 mg/(kg·d)连续皮下注射7 d建立大鼠心肌肥厚模型。造模的第2天起灌胃给药通脉养心丸2.1、1.0、0.5 g/(kg·d),连续给药4 周。超声心动检测大鼠心脏功能和结构的变化,测量大鼠心脏和左心室系数、血清丙二醛(MDA)及脂质过氧化物(LPO)含量、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(郧杂匀原孕曾)活性来判断通脉养心丸对ISO 所致大鼠心肌肥厚的保护作用。[结果] 模型组大鼠室间隔厚度(IVID)和左室内径(IVS)值均不同程度增大,模型组射血分数(EF%)和短轴缩窄率(FS%)显著降低,心脏和左心室系数显著增加,血清SOD 和GSH-Px 的活性显著降低,血清MDA和LPO 含量显著升高。通脉养心丸治疗4周后,大鼠心脏EF%、FS%值和血清SOD活性不同程度升高,MDA 和LPO 含量不同程度降低,通脉养心丸2.0、1.0 g/kg 不同程度升高GSH-Px 活性。[结论] 通脉养心丸对ISO 诱导的大鼠心肌肥厚具有保护作用,其作用机制可能与提高大鼠的抗氧化能力有关,提高大鼠的抗氧化功能可能是通脉养心丸抗心肌肥厚的机制之一。

Abstract:
[Objective] To explore the effects of Tongmai Yangxin pills on myocardial hypertrophy caused by isopropylarterenol(ISO) in rats. [Methods] The myocardial hypertrophy was reproduced by hypodermic injection of ISO, 5 mg/(kg·d)for 7 days at back skin in rats. Tongmai Yangxin pills 2.0, 1.0, 0.5 g/(kg·d) were administrated by gavaging from the second day for total 4 weeks. After the last ad-ministration, echocardiography was performed to detect the changes in cardiac structure and function in rats, calculate the quality and the indexes of whole heart and left ventricle, detect the superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activity, malondi-aldehyde(MDA) and lipid peroxide (LPO) concentration in serum. [Results] In the model group the values of septal thickness (IVID) and left ventricular internal diameter (IVS) were increased in varying degrees, the ejection fraction (EF%) and minor narrowing rate (FS%) were significantly lower, left ventricular heart coefficient was significantly increased, serum SOD and GSH-Px activity were significantly reduced, the serum levels of MDA and LPO were significantly increased 4 weeks after treatment by Yongmai Yangxin pills. The value of FS%, EF% and SOD activity were increased in varying degrees, the MDA and LPO were reduced in varying degrees. Tongmai Yangxin pills at 2.0, 1.0 g/kg could elevate GSH-Px activity in varying degrees. [Conclusion] Tongmai Yangxin pill has a protective effect against myocardial hypertrophy induced by ISO in rats through enhancing the anti-oxidative activity.
