[目的] 研究化浊行血颗粒对食饵性动脉粥样硬化(AS)模型大鼠颈总动脉过氧化物酶体增殖物激活受体(PPARs)、核因子-κB(NF-κB)的影响,探讨中药治疗AS作用机制。[方法] 实验动物按体质量随机分为正常对照组、模型组和药物组,每组12只,采用高脂饲料法建立大鼠食饵性AS模型。药物组给予化浊行血颗粒水溶液,正常对照组、模型组分别给予等容量的蒸馏水,连续8周。实时定量PCR测定PPARmRNA,免疫印迹法观察NF-κB表达。[结果] 与正常对照组比较,模型组颈动脉内PPARα、PPARβ及PPARγ含量显着降低(P<0.01),药物组PPARα、PPARβ及PPARγ含量显着升高(P<0.01),模型组和药物组NF-κB均显着增高(P<0.01);与模型组比较,药物组PPARα、PPARβ及PPARγ含量显着升高(P<0.01),药物组NF-κB显着降低(P<0.01).[结论] 化浊行血颗粒可激活PPARα、PPARβ及PPARγ,拮抗NF-κB,从而抑制AS的发生、发展。 |

Effect of Huazhuo Xingxue grains on inflammatory factor in the common carotid artery of atherosclerosis rats |

[Objective] To explore the effect of Huazhuo Xingxue grains on peroxisome proliferator-activated receptor (PPAR) and nuclear factor kappa B (NF-κB) in the common carotid artery of atherosclerosis rats. [Methods] The rats were randomly divided into control, model, and medicine groups. The medicine group was administrated with Huazhuo Xingxue grains for a course of 8 weeks after the atherosclerosis model rats were established by high fat diet, the control group and model group were taken oral equivolumetric distilled water. PPAR mRNA was measured by real time PCR, NF-κB was measured by western blotting. [Results] The PPARα, PPARβ and PPARγ of model group obviously decreased, NF-κB of model, medicine groups obviously increased, which were significantly difference with the control group (P<0.01). The PPARα, PPARβ and PPARγ of medicine group obviously increased, which were significantly difference with the model group (P<0.01). NF-κB of medicine group was significantly less than those of model group(P<0.01). [Conclusion] Huazhuo Xingxue grains can active PPARα, PPARβ and PPARγ and resist NF-κB to inhibit the development of atherosclerosis. |
