[目的] 探讨中药莱菔承气汤对重症急性胰腺炎(SAP)大鼠肾脏细胞凋亡及凋亡基因Bax和Bcl-2表达的影响。[方法] 健康Wistar大鼠72只,随机分为空白对照组(24只)、SAP模型组(24只)和莱菔承气汤治疗组(24只).3组大鼠均于术后6 h,12 h,24 h心脏采血,检测血肌酐(Cr)、血尿素氮(BUN).Annexin V- FITC/PI双染后流式细胞术检测肾脏细胞凋亡指数,RT-PCR检测肾脏组织Bax、Bcl-2 mRNA的表达。[结果] 与对照组相比较,随着时间变化,SAP模型组大鼠血肌酐、血尿素氮水平明显升高(P<0.01),肾脏细胞凋亡指数趋于升高,Bax mRNA表达明显升高(P<0.01),Bcl-2 mRNA表达明显下降(P<0.01).与SAP模型组相比较,莱菔承气汤治疗组大鼠血肌酐、血尿素氮水平明显下降(P<0.01),肾脏细胞凋亡指数下降,Bax mRNA表达明显下降(P<0.01),Bcl-2 mRNA表达明显升高(P<0.01).[结论] 中药莱菔承气汤可能通过调节凋亡相关基因,减少肾脏细胞凋亡以减轻肾脏病理损伤,从而改善SAP时并发的急性肾功能障碍。 |

Effect of Laifu Chengqi decoction on the apoptosis of renal cells in rats with severe acute pancreatitis |

[Objective] To investigate the effect of Laifu Chengqi decoction on the apoptosis of renal cells and expression of apoptosis gene Bax and Bcl-2 in rats with severe acute pancreatitis. [Methods] The 72 healthy Wistar rats were randomly divided into control group (24 rats), SAP group (24 rats) and Laifu Chengqi decoction group (24 rats). Heart blood of rats in the three groups was taken at 6 h, 12 h, 24 h after operation to detect serum Cr and BUN. Renal cells stained by Annexin V- FITC/PI were drawn off from kidney to detect apoptosis rate using flow cytometer. The expression of Bax and Bcl-2 mRNA in pancreatic tissue were detected by RT-PCR. [Results] Compared with the control group, with the change of time, serum Cr and BUN in SAP group were increased significantly (P<0.01). Index of renal cell apoptosis was increased gradually. The expression of Bax mRNA was significantly increased(P<0.01). The expression of Bcl-2 mRNA was significantly decreased(P<0.01). Compared with the SAP group, serum Cr and BUN in Laifu Chengqi decoction group were decreased (P<0.01), and index of renal cell apoptosis was decreased. The expression of Bax mRNA was significantly decreased (P<0.01), the expression of Bcl-2 mRNA was significantly increased (P<0.01). [Conclusion] Laifu Chengqi decoction may regulate the apoptosis related gene to decrease the apoptosis of renal cells, thereby reducing renal pathological damage. |
