[目的] 通过观察乌头汤对糖尿病周围神经病变(DPN)大鼠胫神经神经生长因子(NGF)受体、酪氨酸蛋白激酶A(TrkA)和p75神经营养因子受体(p75NTR)表达及血清NGF含量的影响,探索该方改善DPN的可能机制。[方法] Wistar大鼠腹腔注射链尿佐菌素(STZ)建立糖尿病模型,观察乌头汤对胫神经纤维组织TrkA、p75NTR及血清NGF的影响。[结果] 干预前后模型组和乌头汤组均较空白组FBG显着升高(P<0.01),各组内FBG在干预前后无统计学差异(P>0.05);与模型组比较,乌头汤组血清NGF含量明显升高(P<0.05),神经组织p75NTR明显降低(P<0.05).乌头汤组p75NTR免疫组化染色阳性表达程度较模型组明显降低。[结论] 乌头汤改善DPN的作用可能与其能够调节神经纤维p75NTR含量和血清NGF含量有关。 |

Abstract: |
[Objective] To study the effect and mechanism of Aconite Main Tuber Decoction on NGF,TrkA and p75NTR in rats developed diabetic peripheral neuropathy(DPN). [Methods] To set up pathological model of DPN, wistar rats were given streptozotocin (STZ) by intraperitioneal injection. NGF in serum and TrkA, p75NTR in tibial nerve tissue were observed. [Results] Compared with blank control group, FBG were higher in both before and after the intervention groups and Aconite Main Tuber Decoction group (P<0.01). No significant difference in FBG within the groups before and after intervention (P>0.05). Compared with model control group, NGF in serum were higher and p75NTR in tissue were lower in medicinal group(P<0.05). In medicinal group,immunohistochemistry showed a lower degree positive staining compared with model control. [Conclusion] Aconite Main Tuber Decoction could alleviate DPN by regulating p75NTR/TrkA balance and rising NGF concentration. |
