养心汤对慢性心力衰竭模型大鼠心肌AI、LVEF影响的研究

摘要:
[目的] 通过建立慢性心力衰竭(CHF)大鼠模型,观察养心汤对CHF模型大鼠心肌细胞凋亡指数(AI)和左室射血分数(LVEF)的影响。[方法] 选取SD雄性大鼠70只,随机选取10只大鼠作为空白对照组,其余60只大鼠腹腔注射阿霉素每周4 mg/kg,连续6周,6周后全部大鼠予心脏超声测定心功能,以LVEF<60%为CHF模型复制成功。将模型复制成功大鼠按随机数字表法随机分成模型对照组、养心汤组、卡托普利组。模型对照组、空白对照组予等体积的蒸馏水灌胃,养心汤组予养心汤浸膏8.1 g/(kg·d),卡托普利组予卡托普利2.25 mg/(kg·d),连续4周,实验结束后测定大鼠LVEF、AI及心肌组织病理形态学变化。[结果] LVEF降低,心肌细胞水肿变化,肌纤维断裂缺损,核固缩,胞质不均匀,间质炎细胞浸润,差异有统计学意义(P<0.05),与模型对照组比较,养心汤组、卡托普利组大鼠心肌AI降低,LVEF升高,心肌细胞排列较整齐,形态正常,可见横纵纹,渗出减少,可见断裂缺损,差异有统计学意义(P<0.05)。与卡托普利组比较,养心汤组大鼠心肌AI、LVEF、心肌苏木精-伊红(HE)染色,差异无统计学意义(P>0.05)。[结论] 养心汤能降低心力衰竭大鼠心肌AI,增加LVEF、减少心肌细胞损伤,治疗CHF。

Effects on Yangxin decoction of myocardial AI and LVEF of chronic heart failure model rats

Abstract:
[Objective] Through the establishment of chronic heart failure rat model to observe effects on Yangxin decoction of AI and LVEF of chronic heart failure model rats.[Methods] The 70 male SD rats randomly selected 10 rats as control group, and the remaining 60 rats received intraperitoneal injection of adriamycin 4 mg/kg weekly for 6 weeks. After 6 weeks, all the rats measured the heart function, with LVEF<60% for heart failure model was established successfully. The rat model was established successfully were randomly divided into model control group, Yangxin decoction group and captopril group by random numbers. The blank control group, model control group distilled water with equal volume. Yangxin decoction group treated with Yangxin decoction extract 8.1 g/(kg·d) daily. Captopril group with captopril 2.25 mg/(kg·d) daily, administered for 4 weeks. After 4 weeks, all the rats were measured the AI and HE, LVEF staining to observe the myocardial tissue morphological changes.[Results] Compared with the blank control group, model control group rats myocardial AI increased, LVEF decreased, myocardial cell edema, muscle fiber fracture defext, karyopyknosis, uneven cytoplasm, interstitial infiltration of inflammarory cells. The difference was statistically significant (P<0.05). Compared with the model group, decoction group, raise heart myocardial AI captopril group rats decreased, and LVEF increased. Myocardial cells were arranged in order,normal morphology, cisible horizontal and vertical lines, reduce exudation, visible fracture defect. The difference was statistically significant (P<0.05). Compared with the captopril group, Yangxin decoction group rats myocardial AI, LVEF, myocardial HE staining, and there was no significant difference significance(P>0.05).[Conclusion] Yangxin decoction can inhibit chronic heart failure myocardial tissue of rats with AI, increased LVEF, reduce myocardial cell injury, and treat chronic heart failure in heart failure rats.
