活血化瘀法对子宫内膜异位症子宫内膜干细胞的影响

摘要:
[目的] 研究活血化瘀组方姜黄散对子宫内膜干细胞的影响。[方法] 噻唑蓝(MTT)法明确姜黄散及孕三烯酮对人子宫内膜干细胞体外干预的最高无毒浓度及作用时效。对比孕三烯酮阳性对照组及不加药阴性对照组评价姜黄散对子宫内膜干细胞克隆形成、微球体形成能力的影响。将子宫内膜干细胞接种于裸鼠皮下形成异位病灶后予药物干预,对比孕三烯酮阳性对照组及不加药阴性对照组观察姜黄散高、中、低剂量组异位病灶体积及异位腺体数量的变化。[结果] 姜黄散及孕三烯酮对子宫内膜干细胞干预的最高无毒浓度分别为20 mg/mL及10-6 mol/L,且其抑制作用随时间延长而加强。与阴性对照组比较,姜黄散及孕三烯酮可明显抑制子宫内膜干细胞的克隆形成及微球体形成能力。姜黄散高、中剂量及孕三烯酮可明显抑制裸鼠异位病的生长,异位病灶体积及异位腺体数量较给药前均明显减小。[结论] 活血化瘀法具有抑制子宫内膜干细胞增殖的作用。

Abstract:
[Objective] To investigate effect of promoting blood circulation and removing blood stasis on endometriosis endometrial stem cells.[Methods] Research of the treatment of endometriosis by the method of promoting blood circulation and removing blood stasis (Jianghuang San) to endometrial stem cells, which is include clone formation, mammosphere formationand endometrial-like tissue reconstitution in vivo.[Results] Jianghuang San coucld inhibit the proliferation of endometriosis-endometrial stem cellsobviously. There are statistical significance (P<0.05) between test group which added Jianghuang San and control group which added gestrinone to negative group which without any disposal in clone formation and mammosphere formation. The volume and the number of ectopic glands of tumor also have statistical significance (P<0.05) between high and middle group of Jianghuang San and control group to negative group. [Conclusion] Promoting blood circulation and removing blood stasis can prevent the proliferation of endometrial stem cells.
