[目的] 探讨中药一贯煎对子痫前期大鼠胎盘核转录因子-κB mRNA(NF-κB mRNA)和基质金属蛋白酶-9 mRNA(MMP-9 mRNA)表达的影响。[方法] 建立子痫前期Wistar大鼠模型,并灌胃给予中药一贯煎,监测给药前后大鼠血压及24 h尿蛋白变化。PCR法检测孕鼠胎盘NF-κB mRNA及MMP-9 mRNA的表达。[结果] 1)NF-κB mRNA:与正常妊娠组相比,模型对照组及一贯煎治疗组均升高,与模型对照组相比,一贯煎治疗组降低,且差异均有显著性(P<0.05)。2)MMP-9 mRNA:与正常妊娠组相比,模型对照组与一贯煎治疗组均降低,与模型对照组相比,一贯煎治疗组有所升高,且差异均有显著性(P<0.05)。[结论] 一贯煎通过降低子痫前期大鼠胎盘NF-κB mRNA的表达,升高MMP-9 mRNA的表达治疗子痫前期大鼠。 |

[Objective] To explore of the effect of Yiguanjian on expression of NF-κB mRNA and MMP-9 mRNA in placenta of preeclampsia model rat.[Methods] The preeclampsia rat model was established and intragastric administration of traditional Chinese medicine Yiguanjian. The changes of blood pressure and 24 h urine protein of rats were monitored before and after dosing. The PCR method to measure the expression of NF-κB mRNA and MMP-9 mRNA in placenta of pregnant rats.[Results] 1) The expression of NF-κB mRNA in model control group and Yiguanjian treat group was higher compared with the normal group, with statistical significance (P<0.05), and the expression in Yiguanjian treat group was lower compared with model contonl group, with statistical significance (P<0.05). 2) The expression of MMP-9 mRNA in model control group and Yiguanjian treat group was lower compared with the normal group, with statistical significance (P<0.05), and the expression in Yiguanjian treat group was higher compared with model control group, with statistical significance (P<0.05).[Conclusion] Yiguanjian treatments preeclampsia model rat by reducing the expression of NF-k-B mRNA and rising the expression of MMP-9 mRNA in placenta. |
