[目的]筛选对蟾皮华蟾毒配基组分增效减毒的配伍药物并考察该方剂体外对肿瘤细胞及正常细胞的活性影响。[方法]利用析因设计(24)和正交设计L9(34)优化蟾皮华蟾毒配基组分、五味子组分、黄芪组分和甘草酸之间的配伍关系。利用细胞增殖与毒性检测(CCK-8)法检测该方剂对SW620、L02、人细胞因子诱导性杀伤T细胞(CIK细胞)在不同时间点的细胞存活率,并进一步考察其对L02细胞释放丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)的影响。[结果]析因设计确定了4种药物间有交互效应并确定本方剂由4种药味组成;正交设计进一步优选出配伍剂量:蟾皮华蟾毒配基组分2 μg/mL,五味子组分10 μg/mL,黄芪组分5 μg/mL,甘草酸10 μg/mL。组方完成后将该方剂命名蟾五草组分散。与蟾皮华蟾毒配基组分相比,蟾五草组分散能降低SW620细胞存活率,而增加L02、CIK细胞存活率;另外,能降低L02细胞释放ALT。[结论]配伍合理的蟾五草组分散实现了对蟾皮华蟾毒配基组分的增效减毒。 |

Abstract: |
[Objective] To screen compatible herbs of efficacy enhancing and toxicity reducing on Cinobufagin fraction of the skin of Bufo bufo gargarizans Cantor (CFSB) and observe its activity influence to tumorous and normal cell lines in vitro.[Methods] To optimize compatibility about CFSB, Schisandra chinensis fraction, Astragalus fraction and Glycyrrhizic acid by applying Factorial design (24) and Orthogonal design L9(34). Then, to detect the survival rate of SW620, L02, CIK with this Prescription interfered by CCK-8 method in different time. Last, to detect release of the ALT and AST of L02 cell under this Prescription interfered.[Results] Factorial design has determined that four Chinese herbs have the interaction and this Prescription consists of the four Chinese herbs. Further Orthogonal design optimizes the compatibility doses, which is CFSB 2 μg/mL, Schisandra chinensis fraction 10 μg/mL, Astragalus fraction 5 μg/mL and Glycyrrhizic acid 10 μg/mL. This Prescription which is completed combination is named Chanwucao Fraction Powder (CFP). To compare with CFSB, CFP can reduce the survival rate of SW620 and increase the survival rate of L02 and CIK. In addition, it can reduce that L02 release ALT.[Conclusion] CFP which is appropriate combination realizes efficacy enhancing and toxicity reducing on CFSB. |
