[目的]探讨补气益脾组方芪茵荷叶饮对糖耐量异常大鼠糖代谢及骨骼肌NF-κB信号通路的影响。[方法]采用高脂高糖饲料建立糖耐量受损(IGT)大鼠模型,将成模大鼠按体质量随机分为模型组(M)和9组不同剂量配伍组(A、B、C、D、E、F、G、H和I)。采用正交实验设计,选取黄芪、茵陈蒿和荷叶进行三因素三水平实验L9(34)。于干预前及干预6周、12周检测大鼠空腹血糖(FBG)、空腹胰岛素(FINS)及胰岛素抵抗指数(HOMA-IR),观察骨骼肌组织病理变化,实时荧光定量PCR方法检测各组大鼠肌肉组织IκB-α和NF-κBp65基因转录水平。[结果]D组FBG在干预12周时低于给药前水平;与M组比较,各实验组FINS在干预6周、12周(除A和B组)下降明显(P<0.05),HOMA-IR在干预6周时A、D、E、F、G、H组和干预12周时D、F、G、H、I组较M组下降显著(P<0.05)。仅不同剂量的茵陈蒿对FBG影响存在统计学差异,仅不同剂量黄芪对FINS和HOMA-IR调节具有统计学差异,主次顺序为黄芪>茵陈蒿>荷叶。M组大鼠骨骼肌水肿、有炎性细胞浸润,G组NF-κBp65mRNA表达水平较M组明显下降(P<0.05)。[结论]芪茵荷叶饮中黄芪为君、茵陈蒿为臣,辅以荷叶,最佳配伍方案为半量黄芪、2倍量茵陈蒿、半量荷叶。IGT大鼠骨骼肌组织存在炎性反应,G组配伍的芪茵荷叶饮可通过抑制NF-κB信号通路,改善IGT大鼠胰岛素抵抗,预防IGT向2型糖尿病(T2DM)演变。 |

Abstract: |
[Objective] To discuss the effects of Qiyin Heye Decoction on glucose metabolism and NF-κB signaling pathway in skeletal muscle of rats with abnormal glucose tolerance.[Methods] A rat model of impaired glucose tolerance was established by using high-fat and high-sugar diet. The model rats were randomly divided into model control group 1 group and different dose compatibility group 9 groups according to their weight,and a normal control group was set. In this study,orthogonal experiment was used to select three factors of astragalus,capillaris and lotus leaf for three-level experiment L9(34). Fasting blood glucose,fasting insulin and insulin resistance index were measured before and 6 and 12 weeks after intervention. The transcription levels of IκB-α and NF-κBp65 gene in muscle tissue of each group were detected by real-time fluorescent quantitative PCR,and pathological changes of skeletal muscle tissue were observed.[Results] Compared with the model control group,the insulin resistance index was significantly decreased (P<0.05) in group A,D,E,F,G,and H after 6 weeks of administration and in group D,F,G,H,and I after 12 weeks of administration. The insulin resistance index showed a significant difference with time (P<0.01). Rats in the model control group had skeletal muscle edema and inflammatory cell infiltration,but the expression level of NF-κBp65 was significantly decreased compared with the control group (P<0.05).[Conclusion] There was an inflammatory response in skeletal muscle tissue of IGT rats,but the NF-κB signaling pathway was not activated. Qiyin Heye Decoction can improve insulin resistance in IGT rat model and prevent the evolution of IGT to T2DM,but its mechanism needs further study. |
