基于毛蕊花糖苷含量分析陈皮和砂仁在熟地黄炮制中的影响性研究

摘要:
[目的] 针对熟地黄的药性特点,深入研究熟地黄炮制过程中,陈皮与砂仁对其炮制过程中毛蕊花糖苷含量的影响,并建立超高压液相色谱-串联质谱(UPLC-MS)法检测熟地黄中毛蕊花糖苷含量的方法,并根据其含量变化,验证其炮制方法的科学性与合理性,为丰富熟地黄的炮制品种并建立其质量控制标准提供参考。[方法] 参考熟地黄在全国各省市《中药炮制规范》中的炮制方法,采用其中具有临床实用特色的方法,即:陈皮制熟地黄、砂仁制熟地黄(酒蒸法和拌制法),并以2015版《中华人民共和国药典》中熟地黄的质量标准为参照指标,采用UPLC-MS检测技术对其炮制过程中毛蕊花糖苷的含量变化进行研究,并结合性状评分,进行综合评价及统计学分析。[结果] 检测的线性范围为(1~1 000 ng/mL,r=1),其精密度、重复性、稳定性、准确度均较好(RSD<3%)。本实验中炮制后的熟地黄其毛蕊花糖苷的含量均超过药典标准,在炮制过程中当性状达到药典及传统标准时,其毛蕊花糖苷的含量能保持在较高水平,以陈皮制熟地黄后的毛蕊花糖苷含量最高,炮制过程中含量变化最显著。[结论] 陈皮制熟地黄可明显稳定和控制毛蕊花糖苷在熟地黄炮制过程中的下降趋势。这对通过相应的炮制方法,在有效保存熟地黄指标性成分的同时,扩大熟地黄临床使用范围有积极意义。同时UPLC-MS法由于操作简便、速度快、准确度高,可用于熟地黄炮制过程中毛蕊花糖苷含量的检测。

Study on the influence of Tangerine and Amomum on Rehmannia glutinosa-processed on the content of Verbascoside

Abstract:
[Objective] According to the medicinal characteristics of Rehmannia glutinosa,we studied the effects of Tangerine Peel and Amomum on the content of Verbascoside in the process of processing Rehmannia glutinosa. And established UPLC-MS method for detecting the content of Verbascoside in Radix Rehmanniae. According to its content change,the scientificity and rationality of the processing method was verified. It provided reference for enriching the products of Rehmannia glutinosa and establishing its quality control standards.[Methods] We refered to the processing method of Rehmannia glutinosa in the "Chinese Medicine Processing Regulations" of the nation and provinces and adopted the methods with clinical and practical characteristics as follows:the Tangerine Peel-processed Rehmannia glutinosa,and the Amomum-processed Rehmannia glutinosa (wine steaming method and mixing method). Based on 2015 edition of the Chinese Pharmacopoeia,the quality standard of Rehmannia glutinosa was used as a reference index. UPLC-MS was used to detect the content of Verbascoside in the process of processing. Combined with trait scores,the comprehensive evaluation and statistical analysis were operated.[Results] The linear range of detection was (1-1 000 ng/mL,r=1),and its precision,repeatability,stability and accuracy were stable (RSD<3%). The content of Verbascoside in the Rehmannia glutinosa after the experiment was more than the pharmacopoeia standard. When the trait reached the pharmacopoeia or traditional standard in the process of preparation,the content of Verbascoside in the experiment could reach at a high level. The content of the Tangerine Peel processed Rehmannia glutinosa was kept the highest level,and the content change was the most obvious during the processing.[Conclusion] The Tangerine Peel-processed Rehmannia glutinosa can obviously stabilize and control the downward trend of Verbascoside in the processing. Through the corresponding processing methods,it is of great significance to preserve the index components of Radix Rehmanniae and expand the clinical use range of Rehmannia glutinosa effectively. At the same time,UPLC-MS method can be used for the detection of Verbascoside content in the processing of Radix Rehmanniae due to its simple operation,high speed and high accuracy.
