南葶苈子清炒前后高效液相指纹图谱研究

摘要:
[目的] 建立南葶苈子清炒品高效液相(HPLC)指纹图谱,并研究炮制前后指纹图谱的差异。[方法] 采用HPLC法,NUCLEODUR C18色谱柱(4.6 mm×250 mm,5 μm),流动相乙腈(CH3CN)-0.01 mol/L磷酸二氢钠(NaH2PO4)水溶液梯度洗脱,流速1.0 mL/min,柱温30℃,检测波长254 nm,进样量5 μL。[结果] 南葶苈子生品、清炒品HPLC指纹图谱均含有11个共有峰,生品HPLC指纹图谱相似度分析存在一定差异;清炒后南葶苈子样品间差异减小,各批次相似度均在0.89以上,说明相似度良好。清炒品与生品的HPLC指纹图谱差异较大。清炒品共有峰No.23、No.26、No.30、No.35峰面积相对生品均升高,共有峰No.9、No.13、No.14、No.41峰面积均降低。聚类分析结果表明安国、周至、岷县、长治所产南葶苈子,无论生品或清炒品都与其余各省所产南葶苈子存在一定的差异,而清炒炮制能够提高批次间的相似度。[结论] 本研究建立的实验方法可为南葶苈子清炒工艺及其质量标准的制定提供参考。

Study of HPLC fingerprint of Descurainiae semen before and after stir-fry without additional ingredients

Abstract:
[Objective] To establish and compare the fingerprint of the crude and the processed seeds from Deseurainia sophia (L.) Webbex Prantl.[Methods] HPLC method was adopted on NUCLEODUR C18 column (4.6 mm×250 mm,5 μm),with acetonitrile -0.1 mol/L sodium dihydrogen phosphate solution as the mobile phase for gradient elution. The flow rate was 1.0 mL/min,and the column temperature was maintained at 30℃. The detection wavelength was 254 nm,and the injection volume was 5 μL.[Results] There were 11 common peaks in HPLC fingerprint of crude and processed Descurainiae semen. There are differences of the similarity between various samples and the automatically generated reference fingerprints of crude Descurainiae semen. The similarity between various samples and the automatically generated reference fingerprints of processed Descurainiae semen reached above 0.89. There were differences in fingerprint of Descurainiae semen before and after processing. The common peak areas of No. 23,No. 26,No. 30,No. 35 peak were increased significantly in processed Descurainiae semen,but the common peak areas of No. 9,No.13,No.14,No.41 peak were decreased significantly. There were certain differences of Descurainiae semen between Anguo,Zhouzhi,Minxian,Changzhi and the remaining 8 producing areas,whether their crude or processed Descurainiae semen by cluster analysis. The similarity between 12 batch samples were similarityimproved by stir-fry without additional ingredients.[Conclusions] The established method can provide experimental basis for processing technology and quality standards of Descurainiae semen fried without additional ingredients.
