女贞子多糖抗去势小鼠骨质疏松作用研究

摘要:
[目的] 研究女贞子多糖对去卵巢所致小鼠骨质疏松的影响。[方法] 给予去卵巢小鼠女贞子多糖(3 g/kg和6 g/kg,以生药计)8周后取材,用双能射线骨密度仪和微计算机断层扫描技术(micro CT)分析小鼠股骨骨密度和股骨干骺端骨微结构,全自动生化仪检测血浆中钙(Ca)和无机磷(P)水平。体外培养骨髓来源单个核细胞,使用25 μg/L巨噬细胞集落刺激因子(MCSF)和50 μg/L核因子κB受体活化因子配体(RANKL)诱导其向破骨细胞分化,同时给予女贞子多糖(1×10-3、1×10-4、1×10-5 mg/mL,以提取物计)干预处理,研究女贞子多糖对单个核细胞向破骨细胞分化的影响。[结果] 给药8周后,与假手术组相比,模型组小鼠血浆中Ca和P水平显著升高(P<0.05);与模型组相比,雌激素组、3 g/kg和/或6 g/kg女贞子多糖小鼠血浆中Ca和P水平显著下降(P<0.05)。双能射线结果显示,女贞子多糖3 g/kg和6 g/kg组小鼠股骨骨密度均显著高于模型组小鼠股骨骨密度;micro CT结果显示女贞子多糖3 g/kg和6 g/kg组小鼠股骨干骺端骨微结构明显改善。体外研究结果显示,与模型组相比,1×10-3、1×10-4、1×10-5 mg/mL女贞子多糖组的培养上清中TRAP酶活性显著降低(P<0.05),但没有明显剂量依赖关系。[结论] 女贞子多糖可能通过抑制破骨细胞分化及调节钙磷代谢发挥抗骨质疏松作用。

Study on the anti-osteoporosis effect of ligustrum lucidum polysaccharide in ovariectomized mice

Abstract:
[Objective] To study the effect of ligustrum lucidum polysaccharide on osteoporosis in mouse caused by ovariectomized. [Methods] Ligustrum lucidum polysaccharides (3 g/kg and 6 g/kg,calculated as crude drugs) were given to ovariectomized mice for 8 weeks. The bone mineral density and bone microstructure of femurs were measured by DXA and microCT. Calcium and phosphorusinplasma were measured by automatic biochemical instrument. Bone marrow-derived mononuclear cells were cultured in vitro,and 25 μg/L MCSF and 50 μg/L RANKL were used to induce osteoclast differentiation. At the same time,ligustrum lucidum polysaccharides (1×10-3,1×10-4,1×10-5 mg/mL) were used for intervention treatmentto study the effect of ligustrum lucidum polysaccharide on the differentiation of mononuclear cells into osteoclasts. [Results] After 8 weeks of administration,compared with sham group,the plasma levels of calcium and phosphorus in the model group were significantly increased (P<0.05). Compared with the model group,plasma calcium and phosphorus levels inestradiol group,3 g/kg and/or 6 g/kg ligustrum lucidum polysaccharide group were significantly decreased (P<0.05). The results of dual-energy rays showed that the bone mineral density of the femur in the estrogen group,3 g/kg and 6 g/kg ligustrum lucidum polysaccharide groups were significantly increased than that of the model group. The results of micro CT showed that the bone microstructure of the femoral metaphysis of mice in the 3 g/kg and 6 g/kg groups of ligustrum lucidum polysaccharide was significantly improved. In vitro,compared with the model group,the TRAP enzyme activity in the culture supernatant of the 1×10-3,1×10-4,and 1×10-5 mg/mL ligustrum lucidum polysaccharide groupswas significantly reduced (P<0.05),but there was no significant dose-dependent resistant relationship. [Conclusion] Ligustrum lucidum polysaccharide may play an anti-osteoporotic effect by inhibiting osteoclast differentiation and regulating calcium and phosphorus metabolism.
