[目的] 观察经方麻黄附子细辛汤合防己黄芪汤治疗阳离子化牛血清白蛋白(C-BSA)制备膜性肾病小鼠模型的有效性,及对肾组织超氧化物歧化酶(SOD)、丙二醛(MDA)、白细胞介素(IL)-6、单核细胞趋化蛋白-1(MCP-1)水平、核转录因子-κB(NF-κB)表达的影响。[方法] 60只Balb/c小鼠按体质量分层随机分为正常组,模型组,中药低、中、高剂量组各12只。C-BSA制备膜性肾病小鼠模型。给药6周后取血清,全自动生化分析仪检测小鼠血清白蛋白(ALB)、三酰甘油(TG)、总胆固醇(TC)、血肌酐(SCr)、尿素氮(BUN)的水平;苏木精-伊红(HE)染色和过碘酸雪夫(PAS)染色观察肾组织病例形态学变化;酶联免疫吸附(ELISA)试剂盒检测肾脏组织SOD、MDA、IL-6、MCP-1含量表达;运用免疫组化染色法检测肾组织NF-κB蛋白的表达。[结果] 给药6周后,与模型组比较,中药低中高各剂量浓度组TG、TC水平各有不同程度的下降(P<0.05或P<0.01),ALB水平显著提高(P<0.01),小鼠的肾小球和肾小管结构损伤程度较模型组都有了不同程度的改善。肾组织SOD含量均有不同程度的增加(P<0.05或P<0.01),MDA、IL-6、MCP-1含量均明显下降(P<0.01)。小鼠肾组织NF-κB蛋白表达均有不同程度的下降(P<0.05或P<0.01)。[结论] 经方麻黄附子细辛汤联合防己黄芪汤可有效改善脂质代谢紊乱,提高血浆白蛋白水平,减轻膜性肾病小鼠肾脏病理损害,对膜性肾病小鼠具有治疗作用;可有效抑制膜性肾病小鼠肾脏氧化应激反应和炎症反应,其机制可能与抑制NF-κB通路有关。 |

Mechanism study of Mahuang Fuzi Xixin Decoction combined with Fangji Huangqi Decoction on reducing oxidative stress injury in mouse with membranous nephropathy |

Abstract: |
[Objective] To observe the effectiveness of Chinese medicine Mahuang Fuzi Xixin Decoction combined with Fangji Huangqi Decoction in treating mouse model of membranous nephropathy prepared using cationized bovine serum albumin (C-BSA).And its effect on the expression of SOD,MDA,IL-6,MCP-1 and NF-κB in renal tissue.[Methods] The 60 BALB/C mice were randomly divided into normal group,model group,low,middle and high dose group,12 in each.The mouse model of membranous nephropathy was established by C-BSA. Take the serum six weeks after administration.The levels of ALB,TG,TC,SCR and Bun in mice were detected by automatic biochemical analyzer. HE staining and PAS staining were used to observe the morphological changes of renal tissue. The expression of SOD,MDA,IL-6 and MCP-1 in kidney was detected by ELISA system. The expression of NF-κB protein was detected by immunohistochemical staining.[Results] After six weeks of medication,compared with the model group,the levels of TG and TC in low,middle and high dose groups decreased in different degrees (P<0.05 or P<0.01),the levels of ALB are significantly elevated (P<0.01).The structure of the glomerulus and renal tubules of the mice were improved to some extent compared with the model group. The content of SOD in kidney tissue increased in different degree (P<0.05 or P<0.01).The contents of MDA,IL-6 and MCP-1 decreased significantly (P<0.01). The expression of NF-κB protein in the kidney of mice decreased in different degree (P<0.05 or P< 0.01).[Conclusion] Mahuang Fuzi Xixin Decoction combined with Fangji Huangqi Decoction can effectively improve lipid metabolism disorder,increase plasma albumin level,alleviate renal pathological damage in mice with membranous nephropathy,and have therapeutic effect on mice with membranous nephropathy.It can effectively inhibit the oxidative stress and inflammation in the kidney of mice with membranous nephropathy. The mechanism may be related to the inhibition of NF-κB pathway. |
