黄精多糖对妊娠期高血压模型大鼠胎盘组织TGF-β/Smad信号通路的作用研究

摘要:
[目的] 探讨黄精多糖对妊娠期高血压模型大鼠胎盘功能的影响与机制。[方法] 受孕雌性大鼠随机分为对照组、模型组、黄精多糖(低、中、高)剂量组(100、200、400 mg/kg)和硫酸镁组(100 mg/kg),每组10只。除对照组外,其余组大鼠皮下注射L-精氨酸甲酯构建妊娠期高血压模型。检测血压、24 h蛋白尿、胎鼠体质量、胎盘质量;苏木素-伊红(HE)检测胎盘组织病理学;酶联免疫吸附实验(ELISA)检测胎盘组织白介素(IL)-6、IL-10、肿瘤坏死因子-α(TNF-α)含量;Western Blot检测胎盘组织紧密连接蛋白-1(ZO-1)、封闭蛋白(OCLN)、血管内皮生长因子(VEGF)、转化生长因子-β1(TGF-β1)、p-Smad3、Smad3蛋白表达。[结果] 与对照组比较,模型组大鼠血压、24 h尿蛋白、IL-6、TNF-α含量及TGF-β1、p-Smad3蛋白表达显著升高,胎鼠体质量、胎盘质量、IL-10含量及ZO-1、OCLN、VEGF蛋白表达显著降低(P<0.05),且胎盘组织出现病理学损伤。与模型组比较,各给药组大鼠血压、24 h尿蛋白、IL-6、TNF-α含量及TGF-β1、p-Smad3蛋白表达显著降低,胎鼠体质量、胎盘质量、IL-10含量及ZO-1、OCLN、VEGF蛋白表达显著升高(P<0.05),同时胎盘组织病理学损伤减轻。[结论] 黄精多糖可抑制妊娠期高血压模型大鼠胎盘中TGF-β/Smad信号通路激活,改善胎盘功能障碍。

Study on the effect of polygonatum polysaccharides on the TGF-β/Smad signal pathway in placental tissue of pregnancy induced hypertension rats

Abstract:
[Objective] To investigate the impacts and mechanism of polygonatum polysaccharide on placental function in pregnancy induced hypertension model rats. [Methods] Pregnant female rats were randomly grouped into control group,model group,polygonatum polysaccharide(low,medium and high) dose groups(100,200,400 mg/kg) and magnesium sulfate group(100 mg/kg),with 10 rats in each group. In addition to the control group,rats in other groups were subcutaneously injected with L-arginine methyl ester to build the hypertensive model of pregnancy. Blood pressure,24-hour proteinuria,fetal rat body mass and placenta weight were measured;hematoxylin-eosin(HE) was applied to detect the histopathology of placenta;the contents of interleukin(IL)-6,IL-10 and tumor necrosis factor-α(TNF-α) in placenta were detected by enzyme linked immunosorbent assay(ELISA);Western Blot was applied to detect the expression of tight junction protein-1(ZO-1),blocking protein(OCLN),vascular endothelial growth factor(VEGF),transforming growth factor-β1(TGF-β1),p-Smad3 and Smad3 proteins in placenta. [Results] Compared with the control group,the blood pressure,24-hour urine protein,IL-6,TNF-α contents and TGF-β1,p-Smad3 protein expression in the model group were significantly higher;the body weight,placenta weight,IL-10 content and the expression of ZO-1,OCLN and VEGF protein of fetal rats were significantly lower(P<0.05),and pathological damage occurred in placenta tissue. Compared with the model group,the blood pressure,24-hour urine protein,IL-6,TNF-α contents and TGF-β1,p-Smad3 protein expression of rats in administration groups were significantly lower,the body weight,placenta weight,IL-10 content and the expression of ZO-1,OCLN and VEGF protein of fetal rats were significantly higher(P<0.05),and the pathological damage of placenta was alleviated. [Conclusion] Polygonatum polysaccharide can inhibit the activation of TGF-β/Smad signal pathway in the placenta of pregnancy induced hypertension model rats and improve the placental dysfunction.
