[目的] 观察中成药丹龙口服液对小鼠肺间质纤维化的影响,并对比其与西药吡非尼酮作用效果的差异,探讨丹龙口服液干预肺间质纤维化疗效及可能存在的机制。[方法] 通过博来霉素鼻腔滴入法建立肺间质纤维化昆明小鼠模型,造模14 d后开始灌胃给药,治疗28 d。观察小鼠一般情况、肺功能、肺组织形态学染色、羟脯氨酸(HYP)及促纤维化因子水平等。[结果] 丹龙口服液在各指标中均可与吡非尼酮治疗肺间质纤维化呈现相似或更佳水平。在体质量变化及一般情况中中成药较西药治疗更佳,肺功能水平中成药与西药改善情况相似;苏木精-伊红(HE)染色结果显示中成药与西药减轻肺泡炎症结果相似;Masson染色显示西药较中成药更佳;在肺组织HYP及血清、肺泡灌洗液中促纤维化因子表达水平中,两组呈现相似水平。[结论] 丹龙口服液可以对防治小鼠肺间质纤维化中起到一定的作用,其作用与抑制肺组织炎症及降低促纤维化因子水平相关。 |

Effect of Danlong oral liquid on pulmonary tissue lesion in mice with pulmonary fibrosis |

[Objective] To observe the effect of Chinese patent medicine Danlong oral liquid on pulmonary interstitial fibrosis in mice,and compare its effect with that of Western medicine pirfenidone,so as to explore the curative effect of Danlong oral liquid on pulmonary interstitial fibrosis and the possible mechanism. [Methods] The Kunming mouse model of pulmonary interstitial fibrosis was established by bleomycin nasal drip method. The 14 days after the model was made,the mice were given intragastric administration and treated for 28 days. The general condition,lung function,lung histological staining,hydroxyproline (HYP) and profibrotic factor levels of mice were observed. [Results] Danlong oral liquid showed similar or better level than pirfenidone in the treatment of pulmonary interstitial fibrosis. In weight changes and general conditions,the treatment of PCM was better than that of Western medicine,and the improvement of lung function level was similar to that of Western medicine. The results of HE staining showed that the effect of Chinese patent medicine on alveolar inflammation was similar to that of Western medicine. Masson staining showed that western medicine was better than proprietary Chinese medicine. The expression levels of profibrotic factor in HYP in lung tissue,serum and alveolar lavage fluid were similar between the two groups. [Conclusion] Danlong oral liquid can play a certain role in the prevention and treatment of pulmonary interstitial fibrosis in mice,which is related to inhibiting lung tissue inflammation and reducing the level of pro-fibrotic factors. |
