基于代谢组学探讨生脉胶囊对5-氟尿嘧啶化疗小鼠肝脏的保护作用和可能机制

摘要:
[目的] 探讨生脉胶囊对小鼠化疗性肝损伤的保护作用和分子机制。[方法] 比较对照组、化疗组和化疗联合生脉胶囊治疗组小鼠的体质量、肝质量和肝脏指数,检测血清丙氨酸氨基转移酶(ALT)和天冬氨酸氨基转移酶(AST)活性,并对肝脏组织病理形态学进行检查,应用核磁共振氢谱(1H-NMR)代谢组学分析肝脏差异代谢物,探讨生脉胶囊减轻化疗性肝损伤的可能机制。[结果] 化疗联合生脉胶囊显著逆转单纯化疗导致的小鼠体质量、肝质量和肝脏指数的降低,下调血清AST活性,减轻肝组织水肿和嗜酸性样变。代谢组学分析表明,化疗联合生脉胶囊治疗了上调牛磺酸、谷氨酰胺和褪黑素等代谢物的水平,这些差异代谢物主要富集在糖代谢和氨基酸代谢等通路。[结论] 生脉胶囊能够通过调节糖代谢和氨基酸代谢减轻小鼠化疗性肝损伤。

Metabolomic analysis of the protective effect and potential mechanism of Shengmai Capsules on the liver of mice undergoing 5-fluorouracil chemotherapy

Abstract:
[Objective] To explore the protective effect and molecular mechanism of Shengmai Capsules on chemotherapy-induced liver injury of mice. [Methods] The body weight,liver weight and liver index of mice in the control group,chemotherapy group and chemotherapy combined with Shengmai Capsules group were recorded and compared. Serum activities of alanine aminotransferase(ALT) and aspartate aminotransferase(AST) were detected and liver tissues were examined by pathomorphology. Proton nuclear magnetic resonance(1H-NMR) metabolomics were performed to analyze differential metabolites in liver and to investigate the possible role and mechanism of Shengmai Capsules in alleviating chemotherapy-induced liver injury. [Results] chemotherapy combined with Shengmai Capsules treatment significantly reversed the decrease in body weight,liver weight and liver index caused by chemotherapy,and downregulated serum AST activity,as well alleviated edema and eosinophilic changes in liver tissues. Metabolomic analysis showed that combined treatment with chemotherapy and Shengmai Capsule elevates the levels of taurine,glutamine,and melatonin in mice liver. These metabolites were mainly enriched in glucose metabolism pathway and amino acid metabolism pathways. [Conclusion] Shengmai Capsules can alleviate chemotherapy-induced liver injury in mice by regulating glucose metabolism and amino acid metabolism pathways
