健脾化瘀方通过CD44/ERK信号通路抑制肝癌细胞上皮间质转化的作用机制

摘要:
[目的] 观察健脾化瘀方对肝癌细胞上皮间质转化(EMT)及侵袭转移的影响,并探索其作用机制。[方法] 以高转移人肝癌细胞株MHCC97H为研究对象,设立对照组、10%含药血清组、20%含药血清组。含药血清干预24、48 h后,利用划痕实验及Transwell侵袭实验评估细胞迁移及侵袭能力,实时荧光定量聚合酶链式反应(Real-time PCR)及蛋白免疫印记法(Western Blot)检测EMT通路相关基因及蛋白的表达水平。[结果] 与对照组相比,含药血清组的MHCC97H细胞迁移下降,侵袭能力下降;健脾化瘀方含药血清可下调分化群44(CD44)、细胞外信号调节激酶(ERK)、锌指蛋白转录因子(Snail)、波形蛋白(Vimentin)、N-钙黏蛋白(N-cadherin)的基因表达,上调E-钙黏着蛋白(E-cadherin)基因表达。[结论] 健脾化瘀方含药血清可能通过抑制CD44/ERK通路的激活以抑制肝癌MHCC97H细胞EMT及迁移侵袭。

Abstract:
[Objective] To study the inhibiting effect of Jianpi Huayu Decoction(JHD)in epithelial-mesenchymal transition(EMT)of hepatoma carcinoma cell,and to explore the possible mechanism behind it. [Methods] Culture MHCC97H cells in vitro,and divide the MHCC97H cells into three groups according to the stimulations(control group,10% of the drug-containing serum group,20% of the drug-containing serum group).The cell migration and invasion abilities was evaluated by scratch test and Transwell invasion test, and Real-time PCR and Western Blot were used to detect the expression levels of genes and proteins related to the EMT pathway. [Results]Compared with control group,the migration and invasion ability of the drug-containing serum groups were significantly decreased. As well as,the mRNA and protein expression level of Cluster of differentiation 44(CD44),Extracellular regulated kinase (ERK),zinc finger protein transcription factor(Snail),Vimentin and N-cadherin were significantly decreased,while the expression of E-cadherin was increased. [Conclusion] JHD may inhibits EMT,migration,and invasion of hepatocellular carcinoma MHCC97H cells through inhibiting the activation of the CD44/ERK pathway.
