基于UPLC-Q-TOF/MS探究玉屏风袋泡茶治疗桥本甲状腺炎的潜在作用机制

摘要:
[目的] 通过超高效液相色谱串联四级杆飞行时间质谱(UPLC-Q-TOF/MS)与网络药理学探讨玉屏风袋泡茶对桥本甲状腺炎的药效物质基础和作用机制。[方法] 采用UPLC-Q-TOF/MS定性分析玉屏风袋泡茶的化学成分,网络药理学预测玉屏风袋泡茶治疗桥本甲状腺炎潜在靶点,通过动物实验验证玉屏风袋泡茶对桥本甲状腺炎的治疗作用。[结果] 共分析出75个化学成分。网络药理学结果显示,玉屏风袋泡茶治疗桥本甲状腺炎可能与钙离子信号通路、血管内皮生长因子(VEGF)信号通路、Rap1信号通路、白细胞介素-17(IL-17)信号通路以及丝裂原活化蛋白激酶(MAPK)信号通路等有关。动物实验结果显示,玉屏风袋泡茶可以显著降低桥本甲状腺炎模型小鼠的抗甲状腺过氧化物酶抗体(TPOAb)、抗甲状腺球蛋白抗体(TGAb)、促甲状腺素(TSH)水平,显著升高游离三碘甲状腺原氨酸(FT3)、游离四碘甲状腺原氨酸(FT4)水平,维持甲状腺激素及抗体的稳定,保护甲状腺组织内滤泡细胞完整性减少淋巴细胞浸润,降低甲状腺组织IL-17、VEGF、肿瘤坏死因子-α(TNF-α)蛋白表达。[结论] 采用UPLC-Q-TOF/MS 技术,探究玉屏风袋泡茶的药效物质基础,并结合网络药理学和实验验证探究其治疗桥本甲状腺炎潜在作用机制,初步为玉屏风袋泡茶进一步开发利用提供参考依据。

Study on potential action mechanism of Yupingfeng teabags in treatment of Hashintoto’s thyroditis based on UPLC-Q-TOF/MS

Abstract:
[Objective] To explore the regulatory mechanism of Yupingfeng teabags in treatment of Hashintoto’s thyroditis through UPLC-Q-TOF/MS and network pharmacology. [Methods] UPLC-Q-TOF/MS was used to qualitatively analyze the chemical components of Yupingfeng teabags. Network pharmacology was used to predict the potential targets of Yupingfeng teabags in treatment of Hashintoto’s thyroditis. And animal experiments were used to verify the therapeutic effect of Yupingfeng teabags on Hashintoto’s thyroditis. [Results] Totally 75 compounds were identified. The results of Network pharmacology showed that the effect of Yupingfeng teabags on treatment of Hashintoto’s thyroditis might be related to calcium signaling pathway,VEGF signaling pathway,Rap1 signaling pathway,IL-17 signaling pathway,MAPK signaling pathway and other signaling pathways. Animal experiments showed that Yupingfeng teabags significantly reduced the levels of TGAb,TPOAb and TSH in Hashimoto’s thyroiditis model mice,increased the levels of FT3 and FT4,and maintained the stability of thyroid hormones and antibodies. Moreover,Yupingfeng teabags protected the integrity of follicular cells in thyroid tissue,and reduced lymphocyte infiltration,and the expression of IL-17,VEGF and TNF-α protein in thyroid tissue. [Conclusion] The pharmacodynamic basis of Yupingfeng teabags is determined by UPLC-Q-TOF/MS. Its potential mechanism for Hashintoto’s thyroditis is made clear by being combination with network pharmacology and animal experiments,providing reference for the development and utilization of Yupingfeng teabags.
