回神颗粒对急性脑梗死大鼠长时程增强的影响

摘要:
[目的]观察回神颗粒对急性脑梗死模型大鼠神经行为及大鼠海马长时程增强(LTP)的影响,探讨其促进大鼠神经功能和学习记忆能力的机制。[方法]采用Zea-Longa线拴法复制大脑右侧中动脉缺血模型,随机分为对照组、模型组、药物组[回神颗粒,0.7g(/kg·d)],观察回神颗粒治疗对缺血后大鼠神经功能活动,以及大鼠缺血同侧海马内嗅区前穿通纤维-海马齿状回(PP-DG)长时程增强的影响。[结果]未经药物治疗各组测LTP之前的神经功能评分,均较建模后第1天的评分下降,而药物3d组和药物7d组的评分升高,(7d组P<0.05)。缺血同侧海马齿状回兴奋性突触后电位(EPSP)斜率、群体峰电位(PS)幅值均显著降低(P<0.05,P<0.01);与模型组相比,药物组EPSP斜率增大(1d组P>0.05,3d组P<0.05,7d组P<0.01),各组PS幅值均显著升高(P<0.05,P<0.01)。[结论]回神颗粒能改善急性脑缺血大鼠神经行为和学习记忆能力,其机制可能是通过易化大鼠海马齿状回长时程增强效应实现的。

Abstract:
[Objective] To investigate the effects of Hui-shen (Recupera-ting the mind) granule on neurological behavior and long-term potentiation (LTP) of hippocampus in rats with acute cerebral ischemia,and study its promotion mechanism for neurological behavior and the ability of study and memory.[Methods] Rats were randomly divided into control,model and therapy groups.Models of acute cerebral ischemia were established by the method of Zea-Longa line tied.The changes of LTP at perforation pathway and dentate gyrus (PP-DG) were detected.[Results] The neurological function scores of model groups were lower than the scores at the first day after the model was established.The scores of the therapy group on the 3rd,7th day increased (P<0.05).In model groups the slope of excitatory postsynaptic potential (EPSP) and the population spike (PS) height were significantly decreased compared to that in the control group (P<0.05,P<0.01).Compared with the model groups,the slope of EPSP in therapy groups was enhanced (on 1st day P>0.05,on 3rd day P<0.05,on 7th day P<0.01) and the PS height was also enhanced (P<0.05,P<0.01).[Conclusion] Hui-shen granule can improve the neurological behavior,learning and memory ability in rat with acute cerebral ischemia,which may be attributed to the facilitation role on LTP in hippocampal dentate gyrus.
