[目的]探讨整合素连接激酶(ILK)、肿瘤坏死因子α(TNFα)在酒精性肝纤维化大鼠肝组织中的表达及五田保肝液的干预作用。[方法]将100只Wistar大鼠随机分成6组:正常对照组10只,模型组、五田保肝液低、中、高剂量组、易善复组各18只,采用白酒辅以吡唑、玉米油的混合食料灌胃的方法建立大鼠酒精性肝纤维化模型,12周末处死大鼠,以苏木精-伊红(HE)染色观察各组大鼠肝组织纤维化情况,以实时定量反转录-聚合酶链式反应(RT-PCR)法检测大鼠肝组织ILK、TNFαmRNA的表达。[结果]五田保肝液(尤其是中、高剂量)和易善复各组大鼠肝纤维化程度轻于模型组,模型组大鼠肝组织ILK、TNFαmRNA的表达比正常对照组明显升高;五田保肝液和易善复可降低大鼠肝组织ILK、TNFαmRNA的表达,且以五田保肝液中、高剂量效果更显著。[结论]ILK、TNFαmRNA的表达升高可能是大鼠酒精性肝纤维化发生发展的机制之一,而五田保肝液可能通过下调ILK、TNFαmRNA的表达,对大鼠酒精性肝纤维化的发生发展起到抑制作用。 |

[Objective] To investigate the expression of ILK,TNFαmRNA in rats with alcoholic hepatic fibrosis and the intervention effect of Wutianbaoganye.[Methods] 100 Wistar rats were assigned at random into six groups:normal group (10),model control group (18),Wutianbaoganye low dose group (18),middle dose group (18),high dose group (18),Yishanfu group (18).Alcohol hepatic fibrosis was induced in rats by intragastric administration of the mixture of liquor,pyrazol and corn oil.After intervention for 12 weeks,the degree of hepatic fiborsis was observed by HE,and the expression of ILK,TNFαmRNA in liver tissue were detected by RT-PCR.[Results] Compared with rats in model control group,Wutianbaoganye (especially the middle and the high dose) and Yishanfu group could suppress the development of fibrosis and could decrease the expression of ILK,TNFαmRNA.[Conclusion] Wutianbaoganye has an active effect on ameliorating alcohol hepatic fibrosis in rats.The mechanism may be decreasing the expression of ILK,TNFαmRNA to inhibit the occurrence and development of alcohol hepatic fibrosis. |
