针刺后海穴对血管性痴呆大鼠学习记忆能力及海马CA1区突触素蛋白表达和超微结构的影响

摘要:
[目的] 观察针刺后海穴对血管性痴呆大鼠学习记忆功能、海马CA1区突触素蛋白表达及超微结构的影响。[方法] 将48只SD大鼠按随机数字表法随机分为空白组、模型组和针刺组, 每组16只, 采用双侧颈总动脉永久性结扎(2-VO)方法制造血管性痴呆模型, 针刺后海穴3个疗程后, 采用Morris水迷宫、免疫组化及电镜技术观察各组大鼠学习记忆功能、海马CA1区突触素蛋白表达及神经元超微结构的变化。[结果] 与空白组和模型组比较, 针刺组逃避潜伏期、原平台象限停留时间和大鼠海马CA1区突触素蛋白表达差异均有统计学意义(P<0.05);针刺组神经细胞包膜完整, 核内常染色质均匀分布, 线粒体丰富, 线粒体嵴未见明显断裂或缺失, 粗面内质网丰富;神经胶质细胞细胞器较少, 髓鞘未见明显肿胀、撕裂、溶解。[结论] 针刺后海穴可改善血管性痴呆大鼠海马的学习记忆能力, 其机制可能与改善CA1区突触素蛋白表达及超微结构有关。

The effect of the Houhai acupoint acupuncture on learning and memory ability as well as the synaptic protein expression and neuronal ultrastructure of the hippocampus CA1 region in vascular dementia rat

Abstract:
[Objective] To study the effect of Houhai acupoint acupuncture on the learning and memory function of rat which suffers from vascular dementia as well as on the synaptic protein expression and neuronal ultrastructure of hippocampus' CA1 region. [Methods] Forty eight rats were randomly divided into blank group, model group and acupuncture group, Rat VD models established by permanent occlusion of the bilateral common carotid artery, then acupuncture Houhai acupoint for three courses of treatment, Morris water maze was used to assess their learning and memory ability, the change of the hippocampal tissue was studied with immunohistochemistry and transmission electron microscope. [Results] Learning and memory ability scores of acupuncture group, and area synaptic grain protein expression hippocampal CA1 of had the significant difference(P<0.05) compared with blank group. Nerve cell encapsulated, euchromatin in the nucleus were evenly distributed. There were many mitochondria without obvious breakage or missing and abundant rough endoplasmic reticulum;there were a few glial cells, no obvious swelling, tearing and dissolution of myelin. There was micro-filament microtube, abundant mitochondria. [Conclusion] Acupuncture Houhai acupoint can improve vascular dementia rats learning and memory ability, whose mechanism is probably related to improve hippocampal CA1 area synaptic grain protein expression and neurons on ultrastructure.
