补骨脂水提物引起的大鼠肝损害

摘要:
[目的] 观察补骨脂水提物对大鼠的肝损害。[方法] Wistar大鼠连续12周灌胃给予补骨脂及其水提物,取肝脏称质量并计算肝系数,进行血液生化学检测和肝脏组织病理学观察。对补骨脂生药和水提物的高效液相色谱法(HPLC)色谱图进行对比,确定水提物中的主要成分。[结果] 补骨脂生药粉2.10、1.05 g/kg组和水提物2.10 g/kg组大鼠肝脏增大、肝脏系数显着升高。补骨脂生药粉2.10 g/kg组有3/12只大鼠肝脏出现中度弥漫性脂肪变性,补骨脂水提物2.10 g/kg组有2/12只大鼠出现中度弥漫性脂肪变性。补骨脂水提物中主要存在的是补骨脂苷、异补骨脂苷、补骨脂素和异补骨脂素。[结论] 补骨脂水提物可以导致大鼠肝损害。补骨脂苷、异补骨脂苷、补骨脂素、异补骨脂素有肝毒性。

Aqueous Extract of psoralea corylifolia induced liver injury in rats

Abstract:
[Objective] To observe the liver injury induced by aqueous extract of Psoralea corylifolia in rats. [Methods]Rats were intragastrically treated with Psoralea corylifolia and its aqueous extract for 12 weeks. Serum biochemical indices were determined. Liver weight and liver coefficient were measured. Histopathological examination for the liver was also performed with light microscopy. To compared HPLC chromatograms of Psoralea corylifolia and its aqueous extract and to determine the main composition of aqueous extract. [Results] Rats of Psoralea corylifolia 2.10 g/kg, 1. 05 g/kg groups and the aqueous extract 2.10 g/kg group were hepatomegaly, and its liver coefficient increased significantly. There were local diffuse hepatic steatosis in 3 rats of Psoralea corylifolia 2.10 g/kg group and 2 rats of aqueous extract 2.10 g/kg group. Aqueous extract of Psoralea corylifolia mainly contain psoralenoside, isopsoralenoside, psoralen and isopsoralen. [Conclusion] Aqueous extract of Psoralea corylifolia induced liver injury in rats. Psoralenoside, isopsoralenoside, psoralen, isopsoralen have hepatotoxicity.
