[目的] 探讨通络救脑注射液对氧糖剥夺大鼠脑微血管内皮细胞胎盘生长因子(PLGF)及其受体血管内皮生长因子受体-1(VEGFR-1)表达的影响。[方法] 通过氧糖剥夺(OGD)方法复制脑微血管内皮细胞拟缺血损伤模型。实验分为正常对照组、氧糖剥夺模型组、通络救脑注射液干预组,分别采用酶联免疫吸附法和免疫印迹法检测脑微血管内皮细胞条件培养液中PLFG 的含量,以及内皮细胞VEGFR-1 的表达。[结果] 通络救脑注射液可以显著提高氧糖剥夺模型大鼠脑微血管内皮细胞PLGF 及其受体的表达,与模型组相比,差异具有统计学意义(P<0.01)。[结论] 通络救脑注射液能够增加拟缺血损伤大鼠脑微血管内皮细胞PLGF 及其受体的表达,提示提高PLGF 及其受体的表达是该中药复方发挥清热、解毒、通络药效的分子途径之一。 |

Effect of Tongluo Jiunao injection on expression of placental growth factor of brain microvascular endothelial cells in rats injured by oxygen-glucose deprivation |

Abstract: |
[Objective] To observe the effects of Tongluo Jiunao injection on the expression of the placental growth factor (PLGF) and the vascular endothelial growth factor receptor-1(VEGFR-1) in brain microvascular endothelial cells of rats injured by oxygen-glucose deprivation.[Methods] The model of oxygen-glucose deprivation (OGD) was established in the primarily cultured brain microvascular endothelial cells. The endothelial cells were divided into three groups: normal control group, OGD model group and Tongluo Jiunao injection-treated group. The PLGF content of endothelial cells in conditioned media was tested by ELISA. The expression of VEGFR-1 protein in endothelial cells were measured by western blotting. [Results] The expressions of PLGF content and VEGFR-1 were significantly higher after treating with Tongluo Jiunao injection when compared with the OGD model group (P<0.01). [Conclusion] This traditional Chinese medicinal preparation can enhance the expression of PLGF and VEGFR-1 of endothelial cells in rats injured with OGD, suggesting the therapeutic potential of Tongluo Jiunao injection for ischemic cerebral vascular disease. |
