[目的]研究和营安心方对过氧化氢(H2O2)所致大鼠胚胎心肌细胞氧化应激损伤的影响。[方法]用70%乙醇冷凝回流提取和营安心方,采用H2O2建立大鼠胚胎心肌细胞氧化应激损伤模型。实验分为5组,分别为:正常对照组、H2O2(200 μmol/L)模型组、和营安心方高、中、低剂量组,其中和营安心方高、中、低剂量组分别加入和营安心方提取物500、250、125 mg/kg ,24 h之后加入与模型组同浓度的H2O2(200 μmol/L),共同作用2 h.观察心肌细胞四甲基偶氮唑盐(MTT)代谢率、乳酸脱氢酶(LDH)含量、丙二醛(MDA)含量及各组细胞培养液中过氧化氢酶(CAT)和超氧化物歧化酶(SOD)的含量。[结果]H2O2组MTT代谢率显着性降低(P<0.01),LDH及MDA含量显着性升高(P<0.01),CAT及SOD含量显着性降低(P>0.05);和营安心方可使H2O2损伤的心肌细胞MTT 代谢率提高,MDA 含量减少(P>0.05),LDH 活性降低(P>0.05),并且显着性提高心肌细胞的CAT和SOD含量(P>0.05).[结论]和营安心方对受损伤的心肌细胞具有明显的保护作用,其机制可能与抑制氧化损伤有关。 |

Effect of Heying Anxin prescription on oxidative stress injury in embryo myocardium of rats |

Abstract: |
[Objective] To investigate the effect of Heying Anxin prescription on embryo myocardium injury caused by H2O2.[Methods] Reflux condenser extraction with 70% ethanol of Heying Anxin prescription was used and the cardiomyocytes were incubated with H2O2(200 μmol/L) for 2 hours in model group. the same dose of H2O2 was administered for 2 hours after treatment with Heying Anxin prescription (500 mg/kg, 250 mg/kg, 125 mg/kg) in therapeutic group and incubated for another 24 hours. Metabolic rate of MTT, content of MDA, activity of LDH and the levels of CAT and SOD in each group were measured.[Results] Metabolic rate of MTT: compared with control group, metabolic rates of MTT was decreased significantly in H2O2 group(P<0.01), while Heying Anxin prescription had no significant effect on cardiac myocytes of MTT metabolic rates. Content of MDA: compared with that of H2O2 group, the content of MDA was decreased in Heying Anxin prescription groups significantly(P>0.05).Content of LDH: compared with that of control group, the content of LDH was decreased in Heying Anxin prescription groups significantly(P>0.05).Content of CAT: compared with that of H2O2 group, the content of CAT was elevated in Heying Anxin prescription groups significantly(P>0.05).Content of SOD:compared with that of H2O2 group,the content of SOD was elevated in Heying Anxin prescription groups significantly(P<0.01).[Conclusion] Heying Anxin prescription has significant protective effects on injured myocardium cell induced by H2O2 in rats, and it may be related to the inhibition of oxidative damage. |
