[目的] 研究补骨脂酚对人肾上腺皮质细胞(H295R)睾酮及雌二醇分泌的影响。[方法] 实验分为空白对照组及补骨脂酚不同浓度组,分别使用CCK-8法及酶联免疫吸附法(ELISA法)检测补骨脂酚对H295R细胞活力、睾酮和雌二醇分泌的影响;同时采用实时荧光定量聚合酶链式反应法(ReAl-time PCR)检测补骨脂酚对睾酮和雌二醇合成酶CYP17A1和CYP19A1 mRNA表达的影响。[结果] 补骨脂酚对睾酮合成酶CYP17A1 mRNA无显著影响,但能上调雌二醇合成酶CYP19A1 mRNA含量,具有减少睾酮分泌增加雌二醇分泌的作用。[结论] 补骨脂酚促进H295R细胞中雌激素的产生。 |

Effects of bakuchiol on secretion of testosterone and estradiol from H295R cells in human with carcinoma of adrenal cortex |

[Objective] To study the effects of bakuchiol on testosterone and estradiol secretion from H295R cells. [Methods] The researches were divided into control group and bakuchiol groups. The effects of bakuchiol on cellular activites and the secretion of testosterone and estradiol were detected by CCK-8 method and ELISA method. The levels of CYP17A1 and CYP19A1 mRNA were also detected by Real-time PCR.[Results] Bakuchiol could not affect CYP17A1 mRNA leverl but could up-regulate CYP19A1 mRNA. Bakuchiol had the effect of decreasing the secretion of testosterone but increasing the secretion of estradiol. [Conclusion] Bakuchiol can stimulate estradiol secretion from H295R cells. |
