自噬对电针治疗大鼠实验性坐骨神经损伤后神经再生的影响

摘要:
[目的] 研究自噬在电针治疗周围神经再生中的作用。[方法] 通过手术造成大鼠实验性坐骨神经钳夹伤模型,采用电针治疗,自噬抑制剂三甲基腺嘌呤(3-MA)干预等措施,不同时间点进行坐骨神经功能指数(STI)测定和再生神经组织学观察。[结果] 组间比较发现,在实施干预后第7天和第28天,电针联合自噬抑制剂组的SFI比其他各组更差(P<0.05)。组内比较发现,模型组、电针组在第1、2、7天时的SFI无差异,均在第28天时出现统计学差异(P<0.01);而电针联合自噬抑制剂组在第1、2天时的SFI无差异,但在第7、28天时出现统计学差异(P<0.05)且劣于上述各组。另外,再生神经组织的苏木精-伊红(HE)染色切片和镀银染色切片显示,第28天时,电针联合自噬抑制剂组的再生神经组织和神经纤维的连续性和密集程度较其他各组要差。[结论] 电针在坐骨神经功能的长期恢复中发挥较大作用。在相同电针情况下,抑制自噬反应对坐骨神经功能恢复和形态改善造成较大负面影响,说明电针可能通过促进细胞自噬来改善坐骨神经功能及组织形态。

Influence of autophagy on nerve regeneration induced by electric acupuncture treatment in rats with experimental sciatic nerve injury

Abstract:
[Objective] To study the role of autophagy in electric acupuncture treatment of peripheral nerve regeneration.[Methods] The experimental clamped injury of sciatic nerve was induced in rats through operation. Intervention was given with electric acupuncture treatment,autophagy inhibitor 3-MA. After that treatment at different time points the functional indexes of sciatic nerve (SFI) were determined and the histology of regeneration of nerve was observed.[Results] The comparison between groups found that the SFI in electric acupuncture combined with autophagy inhibitor was more less than other groups (P<0.05). The comparison of inter-groups revealed that the SFI in model group and electric acupuncture group at the first, second and seventh day was not significantly different. All of them were become different at twenty-eight day(P<0.01). The level of SFI in electric acupuncture combined with autophagy inhibitor was not different at the first and second day but at the 7th and 28th day become significantly different(P<0.05) and was less than the above groups. In addition,the biopsy of regenerative nerve tissue stained by HE and silver was showed that the continuity and intensity of regenerative nerve tissue and nerve fibers of electric acupuncture + autophagy inhibitor group was inferior to other groups on 28th day.[Conclusion] The electric acupuncture plays a important role in the course of long-term recovery of sciatic nerve functions. Under the same condition of electric acupuncture,the inhibition of autophagy will produce a larger negative effect for the functional recovery and structural improvement of sciatic nerve, suggesting that the functional recovery and structural improvement can be promoted through cellular autophagy.
