益气活血复方对心肌梗死后慢性心力衰竭大鼠心肌能量代谢的影响

摘要:
[目的] 观察益气活血复方对心肌梗死后慢性心力衰竭(CHF)大鼠心肌能量代谢的影响.[方法] 选用SPF级健康雄性SD大鼠80只,随机选取20只为空白对照组,其余60只大鼠采用左冠状动脉前降支结扎法配合力竭式游泳及节食方法复制慢性心力衰竭大鼠模型.将造模成功的大鼠随机分为中药组、西药组及模型组.空白对照组与模型组给予常规等容积蒸馏水灌胃,每日1次,中药组按生药9.2 g/(kg·d)剂量给予益气活血复方灌胃,每日1次,西药组按生药1.5 mg/(kg·d)剂量给予赖诺普利灌胃,每日1次.持续给药28 d后,禁食不禁水12 h,取心肌组织及血浆.酶联免疫吸附法(ELISA)检测B型脑尿钠肽(BNP)浓度,比色法检测三磷酸腺苷(ATP)浓度;实时荧光定量聚合酶链式反应(PCR)和免疫蛋白印迹法检测过氧化物酶体增殖活化受体γ辅助活化因子1α(PGC-1α)基因和蛋白的表达.[结果] 与空白对照组比较,模型组、中药组、西药组PGC-1α mRNA、蛋白表达及ATP浓度降低,BNP浓度升高,组间差异均有统计学意义(P<0.05).与模型组比较,中药组和西药组PGC-1α mRNA、蛋白表达及ATP浓度升高,BNP浓度降低,组间差异均有统计学意义(P<0.05).中药组与西药组PGC-1α mRNA、蛋白表达及ATP浓度、BNP浓度无明显差异,组间差异无统计学意义(P>0.05).[结论] 益气活血复方可通过激活与心肌能量代谢有关的PGC-1α因子,增加线粒体的产能,以此来改善和纠正心力衰竭.

Effects of Yiqi Huoxue prescription on myocardial energy metabolic with CHF rats after myocardial infarction

Abstract:
[Objective] To study the effects of Yiqi Huoxue prescription on chronic heart failure(CHF) rats after myocardial infarction. [Methods] Twenty rats were selected from eighty healthy SD rats as the blank control group, and the rest was used to prepare CHF rat model by coronary artery ligation in combination with exhausted swimming and food reducing. By detecting the heart color doppler ultrasound and observing signs of changes in rats to determine whether building success. The successful molding rats were randomly divided into Chinese medicine group, Western medicine group and model group, blank control group(n=15) and model group(n=14) were given conventional volume distilled water by intragastric administration, 1 time a day, the Chinese medicine group(n=15) was given 9.2 g/(kg·d) crude drugs dose of Yiqi Huoxue prescription every day by intragastric administration, Western medicine group (n=14) was given 1.5 mg/(kg·d) crude drugs dose of Lisinopril every day by intragastric administration. After continuous administration of 28 days, fast and only drink water for 12 hours, getting myocardial tissue and plasma. The concentration of BNP was detected by ELISA, and the concentration of ATP was detected by colorimetric method, using Real-time fluorescent quantitative PCR and immune protein imprinting method to detect PGC-1α gene and protein expression. [Results] Compared with the blank control group, the mRNA expression and the protein expression of PGC-1α and the concentration of ATP were decreased, while the concentration of BNP was increased in model, Chinese medicine and Western medicine group, showing statistical difference (P<0.05). Compared with the model group, the mRNA expression and the protein expression of PGC-1α and the concentration of ATP were decreased, while the concentration of BNP was increased in Chinese medicine and Western medicine group, showing statistical difference (P<0.05). Compared Chinese medicine group with the Western medicine group, the mRNA expression and the protein expression of PGC-1α and the concentration of ATP and the concentration of BNP had no obvious difference, showing there was no statistically significant difference between the groups(P>0.05). [Conclusion] Yiqi Huoxue prescription can activate the PGC-1α factor which is related to myocardial energy metabolism, increase the capacity of mitochondria, in order to improve and correct heart failure.
