[目的] 研究心脑舒通对缺氧/复氧损伤神经细胞的保护作用及其机制。[方法] 体外培养 Neuro-2a 细胞, 分为正常对照组、缺氧/复氧损伤模型组、心脑舒通(10、25、50 mg/L)给药组, 建立缺氧缺糖(OGD)4 h/复氧复糖(Rep)24 h损伤模型, 检测心脑舒通对各组细胞增殖活力、乳酸脱氢酶(LDH)漏出量、活性氧(ROS)水平和线粒体膜电位(△Ψm)的影响。[结果] 与缺氧/复氧组比较, 心脑舒通可以明显增加Neuro-2a细胞活力, 减少LDH漏出, 降低ROS水平, 增强线粒体膜电位。[结论] 心脑舒通对缺氧/复氧损伤神经细胞具有明显保护作用, 其机制与其影响活性氧水平和线粒体膜电位有关。 |

[Objective]To study the protective effect of Xinnao Shutong on hypoxia/reoxygenation injured nerve cells and its mechanism. [Methods] Neuro-2a cells were cultured in vitro, divided into control, model, low-dose, mid-dose and high-dose Xinnao Shutong (10、25、50 g/mL) groups. Establishmenting OGD 4 h/Rep 24 h damage model, detecting cell viability, lactate dehydrogenase (LDH) leakage, reactive oxygen species(ROS)level and mitochondrial membrane potential (△Ψm). [Results] Compared with model group Xinnao Shutong can significantly increase the Neuro-2a cell activity, reduce the leakage of LDH, decrease the level of ROS, enhance the mitochondrial membrane potential. [Conclusion] Xinnao Shutong has obvious protective effect on hypoxia/reoxygenation injured nerve cells, and the mechanism is related to influence ROS level and mitochondrial membrane potential on Neuro-2a. |
