参草通脉颗粒影响慢性心力衰竭大鼠血栓形成的实验研究

摘要:
[目的]观察参草通脉颗粒对心力衰竭大鼠血栓形成的影响。[方法]选取80只大鼠,随机取20只为正常对照组,其余大鼠经冠脉结扎法配合力竭式游泳、节食等方法造成慢性心力衰竭模型,将造模成功的大鼠分为模型组、中药治疗组、西药治疗组,共4组,均以灌胃给药或蒸馏水28 d。利用蛋白免疫印迹法测定各组血栓素B2(TXB2)、降纤酶原抑制物(PAI-Ⅰ)的变化情况及血清脑钠素(BNP)的浓度,用比色法测定高能磷酸盐(ATP)浓度。[结果]与正常对照组相比,模型组及各组大鼠TXB2、PAI-Ⅰ及BNP含量显著升高(P<0.01),ATP水平显著降低(P<0.01);与模型组相比,中药治疗组和西药治疗组TXB2、PAI-Ⅰ及BNP含量明显降低(P<0.01),ATP明显升高(P<0.01);西药治疗组与中药治疗组血清中TXB2、PAI-Ⅰ明显降低(P<0.01),ATP和BNP含量无明显差异(P>0.05)。[结论]中药复方参草通脉颗粒可显著降低心力衰竭大鼠BNP的含量水平,降低血小板活化作用,减少心力衰竭后血栓的发生,能够改善线粒体能量代谢,改善心功能,抑制或逆转心室重构。

Experimental study on the effect of Shencao Tongmai granule on thrombosis in chronic heart failure rats

Abstract:
[Objective] To observe the effect of the pulse of the herb on thrombosis in heart failure rats.[Methods] A total of 80 rats, twenty were randomly selected as the normal control group, the remaining rats by coronary artery ligation method with the exhaustive swimming and diet to make chronic heart failure model, the successful model of the rats were divided into model group, treatment group of Chinese medicine, western medicine treatment group, a total of 4 groups, all with intragastric administration or distilled water for 28 days. Thromboxane B2 were measured by Western blot assay (TXB2), fibrinolytic proenzyme inhibitor (PAI-Ⅰ) and the changes of serum brain natriuretic peptide (brain natriuretic peptide, BNP) for the determination of the concentration of high energy phosphate Colorimetry (adenosine triphosphate, ATP) concentration.[Results] Compared with the normal group, model group and the other groups of rats TXB2, PAI-Ⅰ and BNP levels increased significantly (P<0.01), ATP level were significantly decreased (P<0.01); compared with the model group, Chinese medicine group and Western medicine group TXB2, PAI-Ⅰand BNP content decreased significantly (P<0.01). ATP decreased significantly (P<0.01); TXB2, PAI-Ⅰ of serum in medicine group and Chinese medicine group decreased significantly (P<0.01), no significant difference between the ATP and BNP content (P>0.05).[Conclusion] Shencao Tongmai granule of the traditional Chinese medicine compound can significantly decrease the level of BNP content in rats with heart failure, reduce platelet activation, reduce the incidence of thrombosis after heart failure, can improve mitochondrial energy metabolism, improve heart function, inhibiting or reversing ventricular remodeling.
