护场理论指导下的箍围法对皮肤脓肿大鼠血浆IL-10、IL-1β的影响 |

摘要: |
[目的] 观察箍围药红肿消外用对皮肤脓肿大鼠血浆内白介素-10(IL-10)、白介素-1β(IL-1β)表达的影响。[方法] 40只大鼠,分为空白组、模型组、对照组、预治疗组、治疗组。除空白组外,其余4组皮下注射金黄色葡萄球菌造成皮下脓肿模型。模型组生理盐水换药,对照组莫匹罗星软膏外涂换药,预治疗组在注射细菌后4 h红肿消2 mL换药,治疗组红肿消2 mL换药,药物范围超过肿胀范围1 cm,换药每日1次。分别于治疗3、7、14、18 d心脏取血2 mL,酶联免疫吸附(ELISA)法检测血浆内IL-10、IL-1β的表达。[结果] 治疗组在治疗后第3天的IL-10含量与空白组比较都呈增加趋势,与空白组相比,对照组、治疗组IL-10的含量差异有统计学意义(P<0.01或P<0.05)。各组IL-1β含量与空白组相比呈升高趋势,与空白组IL-1β含量相比,模型组、对照组、预治疗组、治疗组含量差异有统计学意义(P<0.01)。治疗7 d时,各组的IL-10含量呈下降趋势,对照组、预治疗组数值下降明显,预治疗组、治疗组IL-10含量与空白组、模型组、对照组相比差异有统计学意义(P<0.01或P<0.05)。模型组IL-1β含量持续升高,对照组、预治疗组、治疗组IL-1β含量呈下降趋势接近正常水平,与空白组相比,模型组IL-1β含量有统计学意义(P<0.01)。与模型组相比,对照组、预治疗组、治疗组IL-1β含量有统计学意义(P<0.01)。治疗14、18 d时,预治疗组、治疗组IL-10含量呈下降趋势,模型组呈上升趋势,各组IL-10含量相比差异无统计学意义(P>0.05)。治疗14 d,与空白组相比,模型组、预治疗组IL-1β含量有统计学意义(P<0.01),提示药物干预能抑制IL-1β的表达。与模型组相比,对照组、预治疗组、治疗组IL-1β含量有统计学意义(P<0.01)。与对照组相比,预治疗组IL-1β含量有统计学意义(P<0.05)。与预治疗组相比,治疗组IL-1β含量有统计学意义(P<0.05)。治疗18 d,与空白组相比,模型组IL-1β含量有统计学意义(P<0.01)。与模型组相比,对照组、预治疗组、治疗组IL-1β含量有统计学意义(P<0.01),提示药物干预能抑制IL-1β的表达。[结论] 在肿疡期(第3天)应用箍围法指导下的红肿消和百多邦处理大鼠皮肤脓肿未起到“消散毒邪”的作用。在脓疡期(第7天)与百多邦相比箍围法(红肿消)可促进大鼠血浆内IL-10的表达,红肿消早期应用及形成硬结后干预(2 d)两者无明显差异,百多邦和红肿消均抑制血浆内IL-1β的表达,抑制炎症反应,促进护场形成。在溃疡期(14、18 d)均起到抑制IL-1β的表达,促进创面愈合的作用。 |

Effect of Guwei method on the IL-10 and IL-1β in the plasma of rats with skin abscess under the guidance of huchang theory |

[Objective] Effect of the method of "hoop circumference"(Hongzhongxiao) on the expression of IL-10 and IL-1β in the plasma of rats with skin abscess under the guidance of the theory of "defended field".[Methods] Forty rats were divided into blank group, model group, control group, pretreatment group and treatment group. In addition to the blank group, the other four groups of subcutaneous injection of Staphylococcus aureus caused by subcutaneous abscess model. The model group with normal saline treatment, the control group used Mupirocin Ointment dressing in the treatment group, pretreatment group in the injection of bacteria after 4 h of external Hong zhong xiao 2 mL dressing, the treatment group for external use hongzhong elimination tincture 2 mL dressing, drugs over a range of swelling range 1 cm, dressing was changed 1 times one day. In the treatment of 3 d, 7 d, 14 d, 18 d through the heart to take blood 2 mL, ELISA method to detect the expression of IL-10 and IL-1β in plasma.[Results] The level of IL-10 in each group was increased after 3 d, and there was significant difference between pretreatment group and blank group, suggesting that the early application of Hongzhongxiao increase the content of IL-10 in plasma. Compared with the other three groups, there were significant differences in the levels of IL-10 between pretreatment group and treatment group at the 7th day after the treatment. At the 7th day, the early intervention of the Hongzhongxiao and the later than scleroma formation intervention can increased the plasma IL-10 content. In the late stage of treatment (14~18 d), the limitations of the abscess, the group of plasma IL-10 content of no difference.After 3 days treatment, the levels of IL-1β in the model group, the control group, the pretreatment group and the treatment group were statistically significant compared with the blank group (P<0.01). There was no significant difference between model group, control group, pretreatment group and treatment group in IL-1β level (P>0.05). Compared with the model group, the levels of IL-1β in the pretreatment group and the treatment group were statistically significant(P<0.01).[Conclusion] In the infection stage(3 d), the use of Hongzhongxiao and Mupirocin ointment treatment of rat skin abscess did not play to "eliminate evil" role. In the abscess formation stage(7 d), compared with Mupirocin ointment,hongzhong elimination tincture can promote rat plasma IL-10 expression, inhibition of IL-1β expression, inhibition of inflammation and promote the formation of huchang. |
