补骨脂水提药渣灌胃给药4周的大鼠亚慢性毒性实验研究

摘要:
[目的] 比较补骨脂生药和补骨脂水提后药渣对雌雄大鼠的毒性作用。[方法] 分别将雌、雄SD大鼠按体质量随机各分为5组,每组7只,共70只。补骨脂生药高、低剂量组,药渣高、低剂量组分别为6 g/kg(生药量)和3 g/kg(生药量),灌胃给药4周,进行相关血清生化指标检测,计算脏器系数。测定肝脏组织中总抗氧化能力(T-AOC)和丙二醛(MDA)进行组织病理学检查。[结果] 经过4周灌胃给药,生药组大鼠体重较对照组显著降低,而药渣组体重较对照组没有显著差异;生药组和药渣组的雌、雄大鼠肝、肾脏器系数较对照组均有显著升高;生药组雌、雄大鼠胸腺脏器系数较对照组均显著降低,MDA显著升高,而药渣组高低剂量都无显著差异;雄性大鼠生药高剂量组和药渣高、低剂量组组T-AOC较对照组均有显著升高。与生药组相比,药渣组肝系数、MDA显著降低;而雄性药渣低剂量组T-AOC显著升高。[结论] 与补骨脂生药相比,经水提后的补骨脂药渣的毒性较小。

A four-weeks sub-chronic toxicity study of the debris after decoction of Fructus Psoraleae by intragastric administration in rats

Abstract:
[Objective] Compare the toxicities of crude drug and water extract residues of Fructus Psoraleaein male and female rats.[Methods] Female and male SD rats were randomly divided into 5 groups each according to body weight, with 7 rats in every group. The high and low dose groups of fructus soraleae crude drug, and the high and low dose groups of drug residues were administered by intragastric injection of 6 g/kg or 3 g/kg drug solution, respectively, for 4 weeks. Biochemical indicators were detected and the organ coefficients were calculated. The MDA and T-AOC levels in liver tissues were measured and the tissue damage were detected by histopathological examination.[Results] After 4 weeks of administration, the body weights of the crude drug groups were significantly decreased when compared with the control group. The liver and kidney coefficients both in the crude drug groups and in the residues groups were significantly decreased. In the crude drug groups, the MDA level increased significantly, and the thymus organ coefficient decreased significantly, while no significant difference was found in the residues groups. The T-AOC level in the high-dosedrug administration group and the dregs group of the male rats were significantly elevated compared with the control. Compared with the crude drug group, the liver coefficient and MDA of the dregs group significantly decreased, while the T-AOC of the male dregs low-dose group increased significantly.[Conclusion] The toxicity of Fructus Psoraleae aqueous extract residues is relative smaller than the crude drug.
