基于差示浓度法的远志及其炮制品中三种成分的分析

摘要:
[目的] 采用差示浓度法分析远志及其炮制品中远志(口山)酮Ⅲ(PXⅢ)、3,6'-二芥子酰基蔗糖(DISS)、甘草酸(GA)的含量,以GA为标志物区分制远志与生远志、蜜远志。[方法] 采用超高效液相色谱法(UPLC),以甲醇-0.1%甲酸水为流动相,梯度洗脱,流速0.3 mL/min;柱温60℃;检测波长320、237 nm;进样量2 μL;测定高浓度供试品溶液中低含量成分(GA),测定低浓度供试品溶液中高含量成分(PXⅢ、DISS)。[结果] 差示浓度法能够分析远志及其炮制品中含量悬殊的PXⅢ、DISS和GA的含量。[结论] 该方法灵敏度高、准确性好,以GA为标志物能够区分制远志与生远志、蜜远志。

Analysis of three components in Radix Polygalae and its processed products by the method of differential concentration

Abstract:
[Objective] To establish the method of differential concentration for determining the content of polygalaxanthone Ⅲ (PXⅢ), 3, 6'-disinapoyl sucrose (DISS) and glycyrrhizic acid (GA) in Radix Polygalae and its processed products. GA was used as the marker to distinguish Polygalae Radix processed by gancao decoction from Radix Polygalae (raw) and honey-stir-baked Radix Polygalae.[Methods] UPLC analysis was carried out by the mobile phase of methanol and 0.1% formic acid in a gradient elution mode at 0.3 mL/min. The column temperature was fixed at 60℃. The detection wavelengths were 320 nm and 237 nm. 2 μL sample solution was directly injected for analysis. GA at low-content was detected by the high-concentration sample solution, and PXⅢ, DISS at high-content was detected by the low-concentration sample solution.[Results] The content of PXIII, DISS and GA was simultaneously detected in Radix Polygalae and its processed products by the method of differential concentration.[Conclusion] This method was sensitive and accurate. GA can be used as marker to distinguish Polygalae Radix processed by gancao decoction from Radix Polygalae and honey-stir-baked Radix Polygalae.
