交泰丸对慢性温和不可预知性应激抑郁大鼠肠道菌群的影响

摘要:
[目的] 利用16S rDNA高通量测序技术研究交泰丸对慢性温和不可预知性应激(Chronic Unpredictable Mild Stress,CUMS)抑郁大鼠肠道菌群的影响,探讨交泰丸的抗抑郁作用。[方法] 采用CUMS方法制备抑郁大鼠模型,在实验结束后取各组大鼠粪便,通过16S rDNA高通量测序技术分析其菌群结构变化。[结果] 与空白组相比,模型组的毛螺菌科(Lachnospiraceae)、拟杆菌属(Bacteroides)、布劳特氏菌属(Blautia)丰度下降,瘤胃菌科(Ruminococcaceae)丰度上升;与模型组相比,交泰丸给药组的毛螺菌科(Lachnospiraceae)、拟杆菌(Bacteroides)和阿克曼氏菌(Akkermansia)的丰度提高,而瘤胃菌科(Ruminococcaceae)的丰度降低。[结论] 交泰丸对CUMS模型大鼠具有抗抑郁作用,其作用机制可能与调控肠道菌群,进而改变肠道菌群相关的机体神经递质有关。

Effects of Jiaotai Pills on intestinal flora in depression model mice with chronic unpredictable mild stress

Abstract:
[Objective] Using 16S rDNA high-throughput sequencing technology,the effect of Jiaotai Pills on intestinal flora in rats with chronic unpredictable mild stress(CUMS) was studied,and the antidepressant effect of Jiaotai Pills was discussed. [Methods] The depression rat models were prepared by CUMS method. After the experiment,the feces of each group of rats were collected,and the changes of flora structure were analyzed by 16S rDNA high-throughput sequencing. [Results] Compared with the control group,the abundance of Lachnospiraceae, Bacteroides and Blautia in the model group decreased,and the abundance of Ruminococcaceae increased;compared with the model group,the abundance of Lachnospiraceae,Bacteroides and Akkermansia in Jiaotai Pills group increased,while the abundance of Ruminococcaceae decreased. [Conclusion] Jiaotai Pills has antidepressant effect on CUMS model rats. Its mechanism might be related to the regulation of intestinal flora and the change of neurotransmitters and nerve factors related to intestinal flora.
