[目的] 分析火麻仁生品与炮制品挥发油的化学成分,寻找炮制前后发生显著变化的化学成分。[方法] 采用水蒸气蒸馏法提取火麻仁生品与炮制品的挥发油,利用气相色谱-质谱联用(GC-MS)技术分析其化学成分,并利用内标法计算各成分的相对含量。对炮制前后的成分进行正交偏最小二乘判别(OPLS-DA)分析和配对t检验分析,结合变量权重重要性排序(VIP)值和P值,筛选出显著性差异成分。[结果] 共鉴定出43种化学成分,从火麻仁生品挥发油中鉴定出40种成分,从火麻仁炮制品挥发油中鉴定出36种成分。筛选得到11个显著性差异化合物,其中正己醛、2-戊基呋喃、茴香脑、反,反-2,4-癸二烯醛的含量在炮制后增加,β-榄香烯、α-法呢烯、Δ-杜松烯、芳姜黄酮、植酮、棕榈酸甲酯、亚麻酸甲酯的含量在炮制后降低。[结论] 火麻仁炮制后挥发油的成分组成大致相同,但含量发生较大变化。该研究为火麻仁炮制标准的制定提供了物质基础,为火麻仁的临床应用提供参考。 |

Analysis of volatile oil from raw and processed hemp seed by GC-MS |

[Objective] To analyze the chemical composition of volatile oil from raw and processed hemp seed,and look for chemical components that change significantly before and after processing.[Methods] The volatile oil of raw and processed hemp seed was extracted by steam distillation,its chemical components were analyzed by GC-MS,and the relative contents of each component were calculated by internal standard method. OPLS-DA analysis and pair T test analysis were used to analyze the components before and after processing. VIP value and P value were used to screen out significant difference components.[Results] A total of 43 chemical constituents were identified. 40 compounds were identified from the volatile oil of raw hemp seed and 36 compounds were identified from the volatile oil of processed hemp seed. The 11 compounds with significant differences were screened,among which the contents of hexanal,2-pentylfuran,Anethole,trans,trans-2,4-Decadien-1-al increased after processing,while the contents of β-elemene,α-farnesene,Δ-cadinene,ar-Turmerone,Hexahydrofarnesyl acetone,Methyl palmitate, Methyl linolenate decreased after processing.[Conclusion] The composition of volatile oil was almost the same,but the content of volatile oil changed greatly. This study provided material basis for the formulation of processing standard of hemp seed,and provided reference for clinical application of hemp seed. |
