空间代谢组学在毒性研究领域中的应用进展

摘要:
毒性研究在人类健康领域具有极其重要的地位, 关系着健康风险评估、药物开发、公共卫生和食品安全等方面。代谢组学长期被作为毒性研究的技术手段, 高通量、静态地揭示毒性状态下机体的代谢反应。然而, 器官和组织不同分区的细胞群体构造不同, 其代谢具有异质性, 传统代谢组学技术无法区分代谢物在不同微区的异质性分布。基于质谱成像技术发展起来的空间代谢组学填补了传统代谢组学对原位可视化分析的不足, 它结合了高分辨率的成像技术与高通量的代谢组学分析, 能够实现生物体内代谢物空间分布的精确测定, 给精确定位毒性病灶微区提供了可靠的工具, 为更深入地揭示毒性机制提供了更多可能性。因此, 文章围绕空间代谢组学技术在常见的毒性研究领域——中药毒性、化学药物毒性、农药毒性和慢性环境污染物暴露毒性中的应用展开综述, 其中重点阐述和讨论了空间代谢组学在中药毒性研究中的应用现状并据此分析了当前该技术应用方面的局限性, 以期为推动空间代谢组学在毒性研究领域的应用提供参考。

Advances in the application of spatial metabolomics in toxicity research

Abstract:
Toxicity research holds an extremely significant position in the field of human health, relating to health risk assessment, drug development, public health, and food safety. Metabolomics has long been used as a technical means in toxicity research, revealing the metabolic response of the body in a high-throughput and static manner under toxic conditions. However, different regions of organs and tissues have different cellular compositions, leading to metabolic heterogeneity. Traditional metabolomics techniques cannot distinguish the heterogeneous distribution of metabolites in different micro-regions. Spatial metabolomics, based on the development of mass spectrometry imaging technology, fills the gap traditional metabolomics for in-situ visualization analysis. It combines high-resolution imaging technology with high-throughput metabolomics analysis, enabling precise measurement of the spatial distribution of metabolites within biological bodies. This provides a reliable tool for accurately locating toxic lesions and offers more possibilities for further revealing the mechanisms of toxicity. Therefore, this paper reviews the application of spatial metabolomics technology in the common toxicity research fields of traditional Chinese medicine, chemical drug toxicity, pesticide toxicity and chronic environmental pollutant exposure toxicity, focusing on the application status of spatial metabolomics in the toxicity study of traditional Chinese medicine, and analyzes the limitations of the current application of this technology, in order to provide a reference for promoting the application of spatial metabolomics in the field of toxicity research.
