Inhibitory effect of cinobufagin fraction of the skin of Bufo bufo gargarizans cantor on tumor and normal cell lines in vitro

Abstract
    [Objective] To detect chemical composition of the eighteenth fraction(F18) of toad skin and observe its inhibitory effects on tumor and normal cell lines in vitro. [Methods] First, to test separated F18 by mass spectrometry (MS). Then, to test inhibitory effects of different cell lines (SW480, SW620, BGC823, L02, CIK, C26, CT26, 4T1) with F18 interfered by the MTT method, and to detect the survival rate of SW620, L02, CIK with F18 interfered by CCK-8 method in different time. Last, to detect variation of cell cycle of SW620 with F18 interfered by flow cytometry. [Results] The main component of F18 was cinbufagin, so the F18 was named Cinobufagin Fraction of the Skin of Bufo bufo gargarizans cantor(CFSB). Half-maximal inhibitory concentration values of eight cell lines (SW480、SW620、BGC823、L02、CIK、C26、CT26、4T1) were 1.10、1.15、1.55、4.85、2.63、21.22、31.39、20.75 μg/mL, respectively. The survival rate of SW620, L02 and CIK showed time dependence. The proportion of S and G2/M phase of SW620 cell which was interfered in 2 μg/mL CFSB for 24 h increased from 36.41% to 55.73% and 10.89% to 16.95%, respectively. [Conclusion] CFSB can inhibit the proliferation of tumor and normal cells and also can induce S and G2/M phases cell-cycle arrest in SW620 cell.
