多发梗死痴呆动物模型的改进与评价

摘要:
[目的]对栓子注入法造成多发梗死痴呆(MID)模型加以改进并进行评价。[方法]颈外动脉逆行插管注入血栓建立MID大鼠模型,观察大鼠大脑皮质及海马的病理形态变化,同时应用Morris水迷宫检测MID大鼠的空间学习记忆的变化。[结果]1)病理形态学显示:MID大鼠局部脑区可见多发软化坏死灶,大脑皮质变薄,细胞数目减少,海马区存在广泛的细胞脱失现象,细胞排列稀疏、紊乱。2)行为学检测显示:MID大鼠学习记忆的获得、保持及再学习的能力存在严重障碍,学习记忆的过程延长。[结论]经行为学及形态学的检测,证实了栓子注入法造成多发梗死痴呆模型的可靠性和可重复性。

Improvement and Evaluation on Model of Multiple Infarction Dementia

Abstract:
[Objective] To improve and evaluate the model of multiple infarction dementia (MID) induced by thrombus injection. [Methods] MID was established by inverse injection of thrombus into external carotid artery to observe the pathologic changes in hippocampus and cortex region of brain and the ability of spatial learning and memory with Morris Water Maze(MWM). [Results] The pathologic examination showed multiple infarcted focuses in MID group in brain region. The amount of neural cell decreased in cortex,severe cell lost and cell arrangement disorder in hippocampus region were observed. Behavior test showed that in MID group some severe disturbance existed in the ability of learning acquisition and memory retaining, and the course of learning and memory is delayed. [Conclusions] MID established by thrombus injection was feasible and reproducible.
