[目的]进一步从调节γ-氨基丁酸(GABA)代谢的角度探讨小儿抗痫胶囊的作用机制。[方法]采用高效液相色谱-紫外检测法,观察大鼠大脑皮质、双侧海马的γ-氨基丁酸含量和谷氨酸(GLU)含量的变化。[结果]戊四唑点燃癫痫大鼠脑内GABA的含量降低,GLU含量升高;经小儿抗痫胶囊治疗后,中药高剂量组皮质、海马GABA含量较模型组均有显著升高,中药高剂量组皮质、海马GLU含量均显著低于模型组,表明小儿抗痫胶囊有良好增加模型大鼠脑内GABA含量,降低GLU含量而抗惊厥的作用;而中药高剂量组与低剂量组比较有显著性差异,表明小儿抗痫胶囊对GABA、GLU含量的影响还与剂量显正相关。[结论]小儿抗痫胶囊能显著抑制大鼠戊四唑点燃发作,其疗效与药物剂量呈正相关,作用机制可能与其降低大脑GLU含量,增加大脑GABA含量,影响GABA代谢有关。 |

Abstract: |
[Objective] To observe the change of γ-GABA and Glu content in the brain of epilepsy rats based on the research and explore the effect mechanism of Pedo-anti-epilepsy capsule. [Methods] To observe change of γ-GABA and Glu content in the cerebral cortex and hippocamp of rats by using HPLC and ultra-violet detection method. [Results] The Pedo-anti-epilepsy Capsule had obviously antagonism for pentylenetetrazol lighting and attacking of rats. [Conclusions] The mechanism of action may have related with the change of γ-GABA and GLU content and GABA metabolism in the brain. |
