[目的]观察几种中药成分对人肝癌细胞株Hep-G2糖脂代谢相关基因,包括细胞血管生成素相关生长因子(AGF)、葡萄糖-6-磷酸酶(G-6-Pase)、低密度脂蛋白受体(LDLR)和胰岛素受体底物2(IRS-2)mRNA的影响。[方法]培养人肝癌细胞株Hep-G2,以实时定量逆转录聚合酶链反应(Real Time RT-PCR)法检测中药成分对AGF、G-6-Pase、LDLR和IRS-2 mRNA的影响。[结果]小檗碱可以显著提高Hep-G2细胞AGF和LDLR mRNA的表达,姜黄素可以降低G-6-Pase mRNA的表达。[结论]小檗碱和姜黄素可能通过对糖脂代谢相关基因的调节而对糖尿病有一定的治疗作用。 |

Multi-target screening of traditional Chinese medicine with regulating glucose and lipid metabolism disorder |

Abstract: |
[Objective] To observe the effect of several tradition Chinese medicines on the glucose and lipid metabolism-related genes of human hepatoma cell line Hep-G2,including cell angiopoietin-related growth factor(AGF),glucose-6-phosphatase(G-6-Pase),low density lipoprotein receptor(LDLR) and insulin receptor substrate-2(IRS-2). [Methods] The human hepatoma cell line Hep-G2 was cultured and the impaction of the components of Chinese medicine on AGF,G-6-Pase,LDLR and IRS-2 mRNA expression was detected by real time RT-PCR. [Results] Berberine can improve the AGF and LDLR mRNA expression,and curcumin can reduce the G-6-Pase mRNA expression of Hep-G2 cell line. [Conclusion] The berberine and curcumin have some certain therapeutic effect on diabetes through regulating the related genes with glucose and lipid metabolism. |
