加减地黄饮子预处理对局灶性脑缺血大鼠脑组织NGF表达的影响

摘要:
[目的]观察加减地黄饮子预处理对局灶性脑缺血大鼠脑组织神经生长因子(NGF)表达的影响及其可能的神经元保护机制。[方法]采用栓线法制作大鼠局灶性脑缺血损伤模型(MCAO)。120只Wistar大鼠随机分成6组,每组20只。连续灌胃7d后进行手术操作。苏木-伊红(HE)染色法观察大鼠脑组织病理学改变;免疫组织化学法观察大鼠脑组织NGF的表达。[结果]加减地黄饮子低剂量、高剂量组均能保护梗死区神经细胞,细胞结构较完整;增强梗死区脑组织NGF的表达。[结论]加减地黄饮子预处理可对局灶性脑缺血大鼠脑组织神经元起保护作用,其脑保护作用机制可能与调节大鼠脑组织NGF表达及保护梗死区神经细胞结构有关。

Effect of pretreatment with Dihuang Yinzi decoction on the expression of NGF in rats with focal cerebral ischemia

Abstract:
[Objective] To observe the effect and explore the mechanism of Dihuang Yinzi Decoction (DYD) pretreatment on the expression of NGF in rats with focal cerebral ischemia. [Methods] One hundred and twenty Wistar rats were divided into 6 groups randomly. Focal cerebral ischemia was induced with the middle cerebral artery occlusion methods in rats. The nerve cells’structure and the expression of NGF of brain cerebral Hippocampus in rats with MCAO were observed. [Results] In rats with MCAO DYD pretreatment could increase the expression of NGF and protect the structure of the nerve cell in cerebral Hippocampus. [Conclusion] DYD pretreatment can improve the expression of NGF in rats with MCAO through protecting the nerve cell from the injury of ischemia.
