[目的]对双黄连冻干粉进行血清药物化学分析、阐明其体内药效物质基础,为优化处方配伍提供实验依据。[方法]建立双黄连冻干粉高效液相色谱法(HPLC)指纹图谱,予大鼠尾静脉注射双黄连冻干粉,分别于10min、12h取血,通过空白血清、含药血清、双黄连冻干粉HPLC指纹图谱比对,分析双黄连冻干粉的血清药物成分。[结果]双黄连冻干粉HPLC指纹图谱中分离得到21个化合物,其中7个为已知化合物分别为:绿原酸、连翘酯苷、黄芩苷、连翘苷、木犀草素、汉黄芩苷和黄芩素;给药后10min大鼠血清样品中检测到17个原形成分;12h后可检测到黄芩苷、木犀草素和汉黄芩苷及一未知成分。[结论]7个已知成分可能为双黄连冻干粉体内发挥药效的活性成分及主要物质基础;其中黄芩苷、木犀草素和汉黄芩苷,可能对双黄连冻干粉药效作用的维持发挥了作用。 |

Study on serum active constituents of Shuanghuanglian dry cryogen powder in rats |

[Objective] To investigate the active compounds in Shuanghuanglian Powder Injection (SHL) It was determined by serum phar-macochemistry for illuminating the elemental pharmacodynamic compounds and providing the foundation for optimizing the prescription of SHL. [Methods] Plasma was obtained from Wistar rats 10 min,12 h after intravenous injection of SHL. Based upon the established HPLC fingerprint of SHL,it was compared with HPLC fingerprints of blank serum,serum containing drug and SHL. The serum active compounds in SHL were analyzed. [Results] Twenty-one compounds were separated by HPLC,seven compounds were confirmed to be the SHL respectively as follow:caffeotannic acid,forsythoside,baicalin,forsythin,luteolin,wogonoside and baicalein; seventeen com-pounds contained in SHL were determined in the rats serum after being administrated with SHL for 10 min. Baicalin,luteolin,wogonoside and a unknown compound were determined in the serum after being administrated with SHL for 12 h. [Conclusion] The seven compounds of SHL maybe the main active compounds and the elemental pharmacodynamic compounds of SHL,Baicalin,luteolin,and wogonoside had an important effect in maintaining the curative effect of SHL. |
