胶原蛋白Ⅰ、胶原蛋白Ⅲ在糖尿病大鼠血管病变中的表达及中药的干预

摘要:
[目的] 对中药干预下的糖尿病大鼠动脉损伤模型的胶原蛋白的表达进行研究,为糖尿病大鼠动脉损伤纤维病变学说和中药干预糖尿病血管并发症的可行性提供实验学依据。[方法] 雄性SD大鼠48只,按随机数字表法随机分为A-正常对照组,B-糖尿病对照组,C-中药治疗组,D-二甲双胍治疗组,其中B、C、D 3组为糖尿病模型组,观察各组大鼠股动脉中胶原蛋白Ⅰ、Ⅲ的含量。[结果] 糖尿病模型组与正常对照组比较,胶原蛋白Ⅰ、胶原蛋白Ⅲ均明显升高;各治疗组治疗后与糖尿病对照组比较,胶原蛋白Ⅰ、Ⅲ表达明显下降;中药治疗组与西药治疗组比较,胶原蛋白Ⅰ、Ⅲ表达下降(P<0.05).[结论] 胶原蛋白Ⅰ、Ⅲ在中、晚期糖尿病大鼠的大血管中显着增加,提示糖尿病大血管病变可能与胶原蛋白有关;中药组方加味桃核承气汤能有效降低致纤维化因子胶原蛋白在实验性糖尿病大血管病变中的表达,对糖尿病大血管病变有改善作用。

Expression of collagen protein Ⅰ and Ⅲ in angiopathy of rats with diabetes and intervention with Chinese medicine

Abstract:
[Objective] To study the effect of Chinese medicine on the expression of collagen protein in rats with diabetes and arterial injury for providing the experimental evidences of theory of fiber neuropathy in arterial injury and to explore the intervention feasibility of Chinese medicine on diabetic vascular complications. [Methods] The 48 SD male rats were randomly divided into normal control group(A), diabetic control group (B), Chinese medicine treatment group (C), metformin treatment group (D), to observe the content of collagen protein Ⅰ,Ⅲ in the rats femoral artery. Diabetes was induced in B, C and D group. [Results] Compared with normal group, collagen Ⅰ and collagen Ⅲ were significantly increased in diabetes group; it was significantly decreased after treatment in each group compared with diabetic control group. Compared with Western medicine the expression of collagen Ⅰ, Ⅲ decreased in the Chinese medicine treatment group. [Conclusion] Collagen Ⅰ, Ⅲ increased markedly in vessels of middle, late period of diabetic rats, hinting that diabetic macroangiopathy may be associated with collagen protein; modified Taohe Chengqi decoction can effectively reduce the expression of fibrosis-inducing collagen protein in experimental diabetes induced macroangiopathy and has improvement effect on diabetic macrovascular diseases.
