[目的] 研究松龄血脉康胶囊对肝阳上亢型肾性高血压大鼠的作用机制。[方法] 采用左肾动脉结扎并灌服附子汤的方法复制肝阳上亢型高血压大鼠模型。筛选造模成功的SD大鼠24只,随机分成3组(模型组、中药组、西药组).另以8只正常SD大鼠作为空白组。中药组给予松龄血脉康胶囊,西药组给予雷米普利,空白组和模型组给予生理盐水,连续4周。观察大鼠的外观及易激惹程度,测定痛阈、血压、心脏超声和血浆血管紧张素Ⅱ(AngⅡ)、醛固酮(ALD)含量,检测组织血管紧张素Ⅱ1型受体(AT1R)、血管紧张素转换酶(ACE)、内皮型一氧化氮合酶(eNOS)、内皮素(ET) mRNA表达量。[结果] 肝阳上亢型高血压大鼠造模成功;给药组大鼠症状明显好转,血压下降,血浆AngⅡ、ALD含量降低,组织AT1R、ACE、ET mRNA表达量降低,eNOS mRNA表达量增高;舒张末期左室内径(LVDd)、收缩末期左室内径(LVDs)、舒张末期左室后壁厚度(LVPWd)、收缩末期左室后壁厚度(LVPWs)均显着降低,射血分数(EF)显着升高。[结论] 松龄血脉康能有效改善肝阳上亢症状,通过干预肾素-血管紧张素-醛固酮系统(RAAS)和内皮素-一氧化氮(ET-NO)系统有效地降低血压,进一步抑制心室重构,改善心功能。 |

Abstract: |
[Objective] To research the blood pressure regulating mechanism of Songling Xuemaikang capsule in treating renal hypertension in rats with hyperactivity of liver yang. [Methods] Hypertension in the model group of rats with hyperactivity of liver yang was reproduced by ligaturing the left renal artery and oral infusion with monkshood soup. The 24 SD rats with successful reproduction of hypertension were randomly divided into 3 groups, including model group, Chinese medicine group and Western medicine group. The other 8 normal SD rats were served as blank group. Songling Xuemaikang capsule was given to the rats of Chinese medicine group and Ramipril was given to Western medicine group, while the same volume of saline was fed to the animals in blank group and model group for 4 weeks. The appearance of signs and degree of rats' irritability were observed. The pain threshold, blood pressure, ultrasonic cardiogram, AngⅡand ALD were measured. The expression of AT1R, ACE, eNOS and ET mRNA in tissue were measured. [Results] In the model group these hypertensive rats with hyperactivity of liver yang were reproduced successfully. The Songling Xuemaikang capsule could also improve the irritability symptoms of rats. The increased blood pressure could be inhibited and declined gradually, and the AT1R, ACE, ET mRNA expression were decreased, but the eNOS mRNA was increased after 4 weeks' treatment of Songling Xuemaikang capsule. At the same time, the indicators about LVDd, LVDs, LVPWd and LVPWs were lower. EF was increased significantly. [Conclusion] Songling Xuemaikang capsule can improve the symptoms about hyperactivity of liver yang, reduce the blood pressure by affecting the systems RAAS and ET-NO. In the end the ventricular remodeling is inhibited and the cardiac function is improved. |
