五田保肝液对酒精性肝纤维化大鼠肝组织MMP-2及其抑制剂表达的影响

摘要:
[目的] 研究五田保肝液对酒精性肝纤维化大鼠肝组织中Ⅳ型胶原以及基质金属蛋白酶2(MMP-2)及其抑制因子TIMP-2表达的影响,探讨五田保肝液对大鼠酒精性肝纤维化的治疗效果和可能机制.[方法] 随机将100只Wistar大鼠分成6组:正常组、五田保肝液低、中、高剂量组、模型组、阳性药对照组,其中正常组10只,其他各组均18只.采用白酒加少量玉米油、吡唑的混合物灌胃,制备大鼠酒精性肝纤维化模型,造模12周后处死大鼠,以免疫组化法检测大鼠肝组织Ⅳ型胶原蛋白的表达,实时定量逆转录-聚合酶链反应(RT-PCR)法检测大鼠肝组织MMP-2、TIMP-2mRNA的表达.[结果] 各组Ⅳ型胶原蛋白的表达均比正常组显著升高,但五田保肝液各剂量组和阳性药组Ⅳ型胶原蛋白的表达比模型组降低;模型组大鼠肝组织MMP-2、TIMP-2的mRNA表达比正常组显著升高,但TIMP-2的mRNA表达升高幅度更大;五田保肝液和阳性药组MMP-2mRNA的表达较模型组升高,但TIMP-2mRNA的表达较模型组降低,且五田保肝液以中、高剂量效果更明显.[结论] 五田保肝液可能通过上调MMP-2的表达、下调TIMP-2的表达,从而抑制大鼠酒精性肝纤维化的发生发展.

Effect of Wutian Baogan Ye on expression of MMP-2 and TIMP-2 mRNA of liver tissue in rats with alcohol hepatic fibrosis

Abstract:
[Objective] To investigate the effect of Wutian Baogan Ye on the prevention and treatment of rats with alcohol hepatic fibrosis(AHF)in liver tissue and the possible mechanism. We studied the expression of collagen type IV,MMP-2 and TIMP-2 in liver tissue of rats with AHF. [Methods] The 100 Wistar rats were assigned to six groups randomly: normal group (10), Wutianbaoganye low dose group(18), middle dose group(18), high dose group(18), model control group(18), positive drug group(18). Rat model of AHF was induced by feeding stomach with the mixture of liquor, corn oil and pyrazol. The 12 weeks later, the expression of collagen type IV, MMP-2 and TIMP-2 in liver tissue of rats were observed. [Results] Compared with normal rats, the expression of collagen type IV of rats in all group was increased. Compared with model rats, Wutian Baogan Ye (especially for Wutian Baogan Ye middle, and high dose) and positive drug could decrease the expression of collagen type IV. Compared with normal rats, the expression of MMP-2 and TIMP-2 was increased in model rats, but the TIMP-2 was increased more obviously. Compared with the model rats, the expression of MMP-2 was increased, and the expression of TIMP-2 was decreased in the rats given drug (especially for Wutian Baogan Ye middle and high dose). [Conclusion] Wutian Baogan Ye has an active effect on inhibiting alcohol hepatic fibrosis in rats. Wutian Baogan Ye can increase the expression of MMP-2 and decrease the expression of TIMP-2.
