补肾药与祛风药配伍控制狼疮小鼠狼疮肾炎进展的实验研究

摘要:

[目的] 观察补肾中药与祛风中药配伍对狼疮小鼠病情活动的影响。[方法] 50 只MRL/lpr狼疮小鼠随机分为5组,补肾组、祛风组、补肾祛风组、西药组、生理盐水组,同时设置10 只BAbl/c 小鼠对照。分别给予对应的中西药物治疗6 周,观察小鼠体质量、尿蛋白、尿红细胞、尿白细胞及双链(天然)脱氧核糖核酸(ds-DNA)、白介素-17(IL-17)变化。[结果] 各组小鼠随着周龄增长体质量均有增加,其中祛风组增加较少,与其他各组比较差异有统计学意义(P<0.05)。各给药组ds-DNA 水平低于生理盐水组(P<0.05)。补肾祛风组ds-DNA 下降明显,优于其他各给药组(P<0.05)。与生理盐水组比较,其他各给药组IL-17 下降明显(P<0.05),且补肾祛风组优于西药组,西药组优于补肾组及祛风组(P<0.05)。尿常规情况,生理盐水组7 只小鼠出现红细胞,其他各组与生理盐水组比只数较少,但各组与生理盐水组比较差异均无统计学意义(P>>0.003 12)。生理盐水组所有小鼠均出现尿蛋白阳性,与其他各组比差异有统计学意义(P<0.003 12);除生理盐水组,其他各给药组之间差异无统计学意义(P>>0.003 12)。[结论] 补肾药与祛风药配伍能明显降低狼疮小鼠IL-17 和ds-DNA 表达,起到控制狼疮肾炎的作用,效果优于单独使用补肾药和祛风药。

Abstract:

[Objective] To observe the effects of compatibility of invigorating kidney herbs and dispelling wind herbs used in lupus mice. [Methods] Fifty lupus mice were randomly divided into five groups, and they were invigorating kidney group, dispelling wind group, invigorating kidney and dispelling wind group, medicine group and normal saline group. Another 10 BAbl/c mice were used to compare. Each group was given the corresponding herbs and medicine for six weeks. [Results] The weight of the mice in each group increased with weeks growth, the dispelling wind group increased lower significantly than other groups (P<0.05). The level of ds-DNA in groups that given herbs and medicine was lower than in normal saline group (P<0.05). The level of ds-DNA in invigorating kidney and dispelling wind group declined significantly than other groups (P<0.05). The level of IL-17 in groups that given herbs and medicine declined significantly than normal saline group (P<0.05), and the invigorating kidney and dispelling wind group was lower than medicine group which was lower than invigorating kidney group, dispelling wind group, but no difference among each group. There were seven mice of normal saline group occurred erythrocytes in urine which was more than other groups, but no difference among each group(P>>0.003 12). All the mice of normal saline group appeared urinary protein which were more than other groups (P<0.003 12) and there was no difference between the groups except normal saline group (P>>0.003 12). [Conclusion] Compatibility of invigorating kidney herbs and dispelling wind herbs can control the development of lupus nephritis through the way of declining the expression of IL-17 and ds-DNA in Lupus Mice significantly, which effects were better than using invigorating kidney herbs or dispelling wind herbs alone.
