生肌象皮膏对2型糖尿病大鼠溃疡肉芽组织的Th1/Th2淋巴细胞平衡的影响

摘要:
[目的] 探讨生肌象皮膏对糖尿病大鼠溃疡组织Th1/Th2淋巴细胞平衡的影响。[方法] 将Wistar大鼠随机分为4组,1组为正常对照组,其余3组采用链脲佐菌素(STZ)尾静脉注射制备糖尿病模型,4组动物均于背部进行打孔处理,造成正常对照大鼠溃疡模型与糖尿病溃疡模型。糖尿病溃疡大鼠按照血糖和体质量分层随机分为糖尿病对照组、凡士林组与生肌象皮膏组。观察创面组织白细胞介素(IL)-2、IL-4、IL-10和干扰素-γ(IFN-γ)含量的变化。[结果] 与正常对照组比较,糖尿病对照组大鼠溃疡肉芽组织中IL-2及IFN-γ的表达于实验各个时间点均升高IL-2及IFN-γ的表达于实验各个时间点均升高(P<0.05),而IL-4及 IL-10的含量于实验各个时间点均降低(P<0.05);与糖尿病对照组比较,生肌象皮膏组大鼠溃疡肉芽组织中IL-2及IFN-γ的表达于实验3、7、14、21、30 d均明显降低,而IL-4及 IL-10的表达于实验3、7、14和21天增加(均于实验7 d达到高峰),P<0.05。[结论] 生肌象皮膏促进糖尿病大鼠溃疡创面的愈合与其调节溃疡肉芽组织的Th1/Th2淋巴细胞的平衡有密切联系。

Influence on the balance of Th1/Th2 lymphocyte in the ulcer healing in type 2 diabetic rats treated by the Shengji Xiangpi mastic

Abstract:
[Objective] To investigate the influence on the balance of Th1/Th2 lymphocyte in the ulcer healing in diabetic rats treated by the Shengji Xiangpi mastic(SM). [Methods] Wistar rats were randomly divided into four groups, one group as the normal control group, the remaining three groups rats injected STZ into vena caudalis as the diabetic model. All the four group rats were punched in back, establishing the diabetic ulcer model. The three group rats of diabetic ulcers were randomly divided into diabetic control group, vaseline group and SM group by the blood glucose and body weight. The changes of IL-2、IL-4、IL-10 and IFN-γ in the wound tissue were observed. [Results] Compare with the rats in normal control group,the contents of IL-2 and IFN-γ were higher and the contents of IL-4 and IL-10 were lower at each selected time point in the rats of the diabetic control group during the experiment, and the difference was significant respectively. Compare with the rats in diabetic control group, the contens of IL-2 and IFN-γ were lower on the 3th, 7th, 14th, 21th, 30th day, while the contents of IL-4 and IL-10 on the 3th, 7th, 14th and 21th day were higher in the rats of SM group, and the difference was significant respectively. [Conclusion] The role of Shengji Xiangpi mastic promoting the ulcer healing in diabetic rats, was closely related to regulate the balance of Th1/Th2 lymphocyte.
