蛇床子香豆素类成分的含量测定及指纹图谱研究

摘要:
[目的] 采用高效液相色谱法(HPLC)同时测定不同批次蛇床子中6种香豆素类成分的含量,并建立指纹图谱分析方法评价其质量。[方法] 采用高效液相色谱法测定蛇床子中花椒毒素、异虎耳草素、佛手柑内酯、欧前胡素、蛇床子素与异欧前胡素的含量,并对17批蛇床子药材进行指纹图谱相似度分析。色谱条件为Hedra ODS-2色谱柱(4.6 mm×250 mm,5 μm);甲醇-0.1%甲酸水梯度洗脱,流速0.5 mL/min,检测波长为320 nm.[结果] 测定了不同批次蛇床子中6种化学组分的含量,建立蛇床子HPLC指纹图谱,17批次药材与对照指纹图谱相似度均在0.980以上。[结论] 建立的蛇床子中6种香豆素成分同时测定的HPLC方法及指纹图谱可用于蛇床子的质量控制。

Simultaneous determination of coumarins and fingerprint analysis in Cnidium monnieri (L.) Cuss.

Abstract:
[Objective] A fingerprint analysis and simultaneous determination of six coumarins were established and validated to evaluate the quality of different batches of Cnidium monnieri (L.) Cuss. [Methods] The contents of xanthotoxin, isopimpinelline, bergapten,imperatorin, osthole and isoimperatorin were determined by HPLC. The analysis was carried out on Hedra ODS-2 column(4.6 mm × 250 mm, 5 μm). The mobile phase was methanol-0.1% formic acid water aqueous solution in gradient elution. The flow rate was 0.5 mL/min and the detection wave length was 320 nm. [Results] The contents of six components in various bathes of samples were determined and HPLC fingerprint was established. The similarities of 17 batches of Cnidium monnieri (L.) Cuss. were more than 0.980. [Conclusion] The established method could be suitable to evaluate the quality control of Cnidium monnieri (L.) Cuss.
